Abstract

Pengendalian penyakit tanaman menggunakan agen antagonis lebih ramah lingkungan dibandingkan dengan penggunaan bahan kimia seperti fungisida dan sejenisnya. Salah satu penyakit penting tanaman yang menyebabkan penururan hasil hasil panen yaitu, layu bakteri pada tanaman tomat yang disebabkan oleh patogen Ralstonia solanacearum. Dari beberapa penelitian, bakteri antagonis mampu menghambat patogen Ralstonia solanacearum, yaitu seperti genus Coryneform, Bacillus, Clostridium, Pseudomonas, dan Pantoea. Ditemukan beberapa genus bakteri asal kompos kerbau belang yang dapat menghambat patogen Ralstonia solanacearum. Penelitian dilaksanakan pada bulan Maret sampai dengan bulan Agustus 2018 di Laboratorium Penyakit Tumbuhan Jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Brawijaya, Malang. Tahapan penelitian yaitu : 1) Eksplorasi bakteri dari kompos sapi, 2) Seleksi bakteri dari kompos sapi yang mampu menjadi antagonis Ralstonia solanacearum, 2) Identifikasi bakteri yang mampu menjadi antagonis. Tahap uji antagonis dilakukan secara In vitro menggunakan rancangan acak lengkap (RAL) dengan 4 perlakuan yaitu 3 perlakuan bakteri, dan 1 kontrol. Setiap perlakuan diulang sebanyak 6 ulangan. Variabel yang diamati yaitu, karakterisasi bakteri, dan penghambatan bakteri antagonis terhadap patogen Ralstonia solanacearum. Hasil penelitian didapatkan 3 bakteri yang merupakan bakteri antagonis, dan mampu menghambat patogen Ralstonia solanacearum. Tiga bakteri yang merupakan antagonis yaitu kode S13, S14, dan S18. Uji penghambatan dilakukan secara In vitro, dengan metode pengkabutan. Pengujian antagonis pada media NA menunjukkan 3 bakteri uji menghasilkan zona hambat yang menunjukkan ada penghambatan terhadap bakteri patogen. Dari 3 perlakuan bakteri antagonis, dan 1 kontrol menunjukkan hasil yang sangat berbeda nyata. Zona hambat tertinggi yaitu pada bakteri S13. 3 bakteri antagonis yang ditemukan diidentifikasi untuk menentukan genusnya. Bakteri S13 merupakanii genus Xanthomonas sp, bakteri S18 belum teridentifikasi maka perlu dilakukan uji lanjutan, bakteri S14 merupakan bakteri genus Pantoea sp.

English Abstract

Control of plant diseases using antagonist agents is more environmentally friendly by using chemicals such as fungicides and the like. One important disease that produces yields is organic vegetation in plants guided by the pathogen Ralstonia solanacearum. From several studies, antagonistic bacteria were able to inhibit the pathogen Ralstonia solanacearum, namely genera Coryneform, Bacillus, Clostridium, Pseudomonas, and Pantoea. Found several genera of bacteria from striped buffalo compost that can inhibit the pathogen Ralstonia solanacearum. The study was conducted in March to August 2018 at the Laboratory of Plant Diseases, Department of Plant Pests and Diseases, Faculty of Agriculture, Brawijaya University, Malang. The stages of the research are: 1) Exploration of bacteria from cow compost, 2) Bacterial selection from cow compost which becomes the antagonist of Ralstonia solanacearum, 2) Identification of bacteria that become antagonists. The antagonistic test stage was carried out in vitro using RAW with 4 chambers, namely 3 hours of bacteria, and 1 control. Each treatment was repeated 6 replications. Observing variables are bacterial characterization, and inhibition of antagonistic bacteria against pathogen Ralstonia solanacearum. The results of the study found 3 bacteria which are antagonistic bacteria, and capable of inhibiting the pathogen Ralstonia solanacearum. Three antagonistic bacteria, namely code S13, S14, and S18. The inhibition test was carried out in vitro, by means of fading. Antagonistic testing on NA media showed that 3 test bacteria produced inhibitory zones which showed that there were inhibitions of pathogenic bacteria. Of the 3 oral antagonistic bacteria, and 1 control showed very different results. The highest inhibition zone in S13 bacteria. 3 antagonistic bacteria found to determine the genus. S13 bacteria is the genus of Xanthomonas sp, S18 bacteria have not been identified so it needs to be tested, the S14 bacteria is the genus Pantoea sp.
