Abstract
Semen cair mulai dikembangkan kembali dengan menggunakan pengencer yang ada, baik pengencer konvensional maupun pengencer baru. Semen beku yang sering digunakan untuk IB memiliki kualitas yang lebih rendah dan hanya dapat dipertahankan apabila tersedia Nitrogen cair (N2 cair) secara kontinyu, sedangkan di beberapa daerah di Indonesia N2 cair sulit untuk didapatkan dan berdampak pada rendahnya keberhasilan IB. Sebagai alternatif perbaikan mutu genetik, berbagai pengencer perlu di uji coba untuk menentukan pengencer yang mampu mempertahankan kualitas semen pada suhu penyimpanan 3-50C. Penelitian ini bertujuan untuk membandingkan perbedaan kualitas semen sapi Limousin menggunakan pengencer CEP-2 + 10% kuning telur, Tris Aminomethan + 20% kuning telur dan Skim+ 10% kuning telur selama pendinginan 3-50C. Materi penelitian yang digunakan adalah semen dari pejantan sapi Limousin yang dipelihara oleh Balai Besar Inseminasi Buatan (BBIB) Singosari dengan frekuensi penampungan semen dua kali dalam seminggu menggunakan metode vagina buatan. Semen pejantan Limousin yang digunakan adalah Placid, Uchita dan Kathandra. Lokasi penelitian adalah Laboratorium BBIB Singosari, Malang. Metode yang digunakan dalam penelitian ini adalah metode eksperimental laboratorium dengan tiga perlakuan, yaitu : P0 (CEP-2 + 10% kuning telur), P1 (Tris Aminomethan + 20% kuning telur), P2 (Skim + 10% kuning telur). Masing–masing perlakuan dilakukan 10 kali ulangan dan variable yang diamati, yaitu kualitas Makroskopis (warna, bau, pH, motilitas, viabilitas, abnormalitas dan konsentrasi) dan juga kualitas mikroskopis (% motilitas, % viabilitas, % abnormalitas dan konsentrasi). Analisis statistik menggunakan Rancangan Acak Kelompok (RAK) dan apabila terdapat beda nyata maupun sangat nyata dilanjutkan dengan Duncan Multiple Range Test (DMRT). Hasil analisa ragam menunjukan Tris Aminomethan + 20% kuning telur menunjukan perbedaan yang sangat nyata (P<0,01) meningkat terhadap P0 dan P2 pada semen Sapi Limousin yang disimpan pada suhu 3-5oC pada jam ke-5 dan 24, yaitu P0 (39±3,07%), P1 (42,75±2,55%) dan P2 (36,50±4,32%). Hasil analisis ragam pada viabilitas, Tris Aminomethan + 20% kuning telur juga memberikan perbedaan yang sangat nyata (P<0,01) meningkat atau lebih tinggi daripada pengencer CEP-2 + 10% kuning telur dan Skim + 10% kuning telur pada jam ke-24, yaitu P0 (66,83±1,00%), P1(68,94±1,22%) dan P2 (64,93±0,85%). Abnormalitas pada Tris Aminomethan + 20% kuning telur menunjukan perbedaan yang sangat nyata (P<0,01) menurun atau lebih rendah daripada pengencer CEP-2 + 10% kuning telur dan Skim + 10% kuning telur, yaitu yaitu P0 (19,01±0,46%), P1(17,08±0,76%) dan P2 (19,04±0,50%). Berdasarkan hasil penelitian tersebut, maka dapat disimpulkan bahwa pengencer Tris Aminomethan + 20% kuning telur (P1) menunjukan hasil yang terbaik terhadap kualitas spermatozoa baik motilitas, viabilitas dan abnormalitas selama pendinginan. Ketiga pengencer dapat digunakan pada semen cair dengan lama penyimpanan 24 jam pada suhu 3-50C. Disarankan untuk melakukan penelitian dengan waktu pengamatan yang lebih lama untuk mengetahui penurunan kualitas spermatozoa dan lama kemampuan pengencer dalam mempertahankan motilitas individu spermatozoa.
English Abstract
This study was done to evaluate quality of Limousin’s semen during chilled preservation. The characteristic of spermatozoa were easily damaged, so that they were necessary to protect viability of spermatozoa. Addition of extender was conducted to determine quality and viability of spermatozoa. Each extender were added Egg Yolk as diluents cryopreservation, the treatment were devided in to three groups, they were ; P0 (CEP-2 + 10% Egg Yolk), P1 (Tris Aminomethane + 20% EggYolk), P2 (Skim + 10% Egg Yolk). The result showed that Tris Aminomethan + 20% Egg Yolk gave the best effect on maintaining quality of spermatozoa (motility, viability and abnormality). The motility in three extender at 24h was high significant different (P<0,01), motility increased at Tris Aminomethan +20% egg yolk. Furthemore, the Viability and abnormality showed high significant different (P<0,01) since first time evaluation at 0h until 24h. Tris Aminomethan incresed viability and decreased abnormality from others extenders. Tris Aminomethane showed the best result among others due to the addition of 20% Egg Yolk, as we know that Egg Yolk was an extracellular cryoprotectan to protect and nourish spermatozoa during chilling preservation.
