Abstract
Pengambilan data penelitian dimulai pada bulan Desember 2012 sampai dengan bulan Februari 2013 bertempat di Laboratorium Lapang Sumber Sekar, Fakultas Peternakan, Universitas Brawijaya Malang untuk penampungan semen segar Kambing Boer dan untuk prosesing semen beku di Laboratorium Reproduksi Ternak, Fakultas Peternakan, Universitas Brawijaya. Tujuan penelitian ini adalah mengetahui dan mengkaji pengaruh ekuilibrasi uap nitrogen cair terhadap kualitas semen beku Kambing Boer dengan Glutathione dalam pengencer. Bahan yang digunakan dalam penelitian ini adalah semen segar Kambing Boer umur kurang lebih 2-2,5 tahun dan berat badan 70-80 kg dengan persyaratan motilitas individu minimal 70%. Metode penelitian yang digunakan adalah percobaan laboratorium yang meliputi: Penampungan semen segar Kambing Boer, prosesing semen beku dan analisis data menggunakan uji-t tidak berpasangan SPSS 16.0. Pengenceran semen menggunakan Tris aminomethane Kuning Telur yang mengandung DMSO 7,5% ditambahkan glutathione dengan konsentrasi berbeda. Konsentrasi glutathione yang digunakan adalah P0 = 0 g/100ml, P1 = 0,2 g/100 ml, P2 = 0,4 g/100 ml, P3 = 0,6 g/100 ml. Data Penelitian ini dianalisis menggunakan uji- t tidak berpasangan SPSS 16.0. Data semen setelah pengenceran dan before freezing dianalisis menggunakan Rancangan Acak Kelompok (RAK) dengan 4 perlakuan penambahan konsentrasi gluthathione yang berbeda, yaitu glutathione sebesar (P 0 ) 0 g; (P 1 ) 0,2 g; (P 2 ) 0,4 g; dan (P4) 0,6 g untuk tiap 100 ml pengencer. Post Thawing 4 cm dan 7 cm dianalisis menggunakan uji t - tidak berpasangan.
English Abstract
Encode retrieval research starting in December and ending in February 2013 held at Sumber Sekar Field Laboratory, Faculty of Animal Husbandry, University of Brawijaya for fresh semen shelter Boer Goats. Further processing of frozen semen for Animal Reproduction Laboratory Faculty of Animal Husbandry University of Brawijaya. The purpose of this study was to determine and assess the effect of vapor eq uilibration liquid nitrogen on the quality of frozen semen Boer Goats supplemented Glutathione . The materials used in this study is the fresh semen Boers approximately 2-2.5 years olds and 70-80 kg weight with the requirements of at least 70% individual motility. Semen was diluted in Tris aminomethane medium, containing 10% (v/v) egg yolk and 7,5% Dimethyl sulfoxide (DMSO) in 100ml. Value of the ttest for two independent samples t = 1,223 with 78 degrees of freedom and the significance of these two variables or a P-value (2-tailed) = 0.225> � = 0.05, it can be concluded that the motility of spermatozoa after post thawing with a height of 4 cm and 7 cm were not significantly different (P> .05). Value of the t-test for two independent samples t = -0810 with 78 degrees of freedom and the significance of these two variables or a P-value (2-tailed) = 0.420> � = 0.05, it can be concluded that the viability of spermatozoa after post thawing with altitude 4 cm and 7 cm were not significantly different. Value of the t-test for two independent samples t = -0870 with 78 degrees of freedom and the significance of these two variables or a P-value (2- tailed) = 0387> � = 0.05, it can be concluded that the abnormalities of spermatozoa after post thawing with a height of 4 cm and 7 cm were not significantly different.
