Abstract

Penelitian dilaksanakan di Laboratorium Reproduksi Ternak Fakultas Peternakan Universitas Brawijaya Malang pada bulan Desember 2007 sampai dengan Januari 2008. Penelitian ini bertujuan untuk mengetahui kualitas dan proporsi spermatozoa X dan Y kambing PE hasil sexing dengan metode sentrifugasi dan inkubasi menggunakan 2 gradien BSA. Hasil dari penelitian ini diharapkan dapat digunakan sebagai informasi dan pedoman produksi semen beku kambing PE hasil sexing serta dapat digunakan sebagai pertimbangan bagi peneliti selanjutnya. Materi yang digunakan dalam penelitian meliputi : Semen kambing PE dengan persyaratan motilitas individu minimal 70 persen dan motilitas massa minimal 2 +, Bovine Serum Albumin (BSA) dan pengencer andromed. Metode yang digunakan dalam penelitian ialah metode percobaan menggunakan gradient BSA konsentrasi 5 dan 15 persen. Data hasil penelitian yang diperoleh dianalisis dengan menggunakan analisis ragam Rancangan Acak Kelompok (RAK) dengan 3 perlakuan dan 10 kelompok ulangan, untuk membandingkan masing-masing perlakuan dilanjutkan dengan Uji Jarak Berganda Duncan serta Uji Chi Square untuk membandingklan spermatozoa X dan Y pada semen segar kambing PEHasil penelitian menggunakan gradien BSA Densitas Albumin menghasilkan kualitas tertinggi meliputi motilitas individu sebesar 56,19 ± 1,90 persen pada lapisan 5 perlakuan sentrifugasi, persentase spermatozoa hidup tertinggi sebesar 59,65 ± 4,51 persen pada lapisan 1 perlakuan inkubasi 20 menit, dan konsentrasi tertinggi sebesar 64,90 ± 24,16 juta/ml pada lapisan 5 perlakuan sentrifugasi. Spermatozoa X tertinggi diperoleh pada lapisan 1 perlakuan inkubasi 10 menit sebesar 55,65 ± 4,14 persen, spermatozoa Y tertinggi diperoleh pada lapisan 5 perlakuan inkubasi 20 menit sebesar 62,19 ± 7,27 persen. Kesimpulan yang diperoleh dari hasil penelitian antara lain : Kualitas spermatozoa meliputi motilitas individu mengalami penurunan rata-rata 25,29 persen dari spermatozoa segar dan persentase hidup mengalami penurunan ratarata 38,44 persen. Motilitas individu tertinggi terdapat pada lapisan 5 perlakuan sentrifugasi sebesar 56,19 ± 1,90 persen, persentase spermatozoa hidup tertinggi terdapat pada lapisan 1 perlakuan inkubasi 20 menit sebesar 59,65 ± 4,51 persen, dan konsentrasi spermatozoa tertinggi terdapat pada lapisan 5 perlakuan sentrifugasi sebesar 64,90 ± 24,16 juta/ml. Spermatozoa X tertinggi diperoleh pada lapisan 1 perlakuan inkubasi 10 menit sebesar 55,65 ± 4,14 persen dan Spermatozoa Y tertinggi diperoleh pada lapisan 5 perlakuan inkubasi 20 menit sebesar 62,19 ± 7,27 persen.

English Abstract

The study was carried out in Animal Reproduction Laboratory, Animal Husbandry Faculty from Desember 2007 until January 2008. The aim of this study was to know the quality and proportion of spermatozoa X and Y PE Goat after sexing with sentrifugation and inkubation methods by using two gradien of BSA. It is hoped that the result of this study can be used as an information and orientation to produce freezing semen PE Goat from sexing process also can be used as consideration for the next research. The material of this study were : PE Goat semen high quality which minimum 70 percent individual motility and mass motility 2+, Bovine Serum Albumin (BSA) and Andromed dilution. The study method was experiment method used gradien of BSA with concentration 5 and 15 percent. Furthermore, the data result was analyzed by using cluster Randomized Block Design using 3 treatments and 10 replications, to compare each dilution treatment ang continued with Duncan Double distance test and Chi Square test to compare the X and Y spermatozoa from PE Goat fresh semen. The study result using two gradiens of BSA Albumin Density showed the highest quality of individual motility was 56,19 ± 1,90 percent were fifth layer on sentrifugation treatment, viability 59,65 ± 4,51 percent were first layer on inkubation 20 minutes treatment, and highest concentration was 64,90 ± 24,16 million/ml on fifth layer on sentrifugation treatment. The highest proportion of X spermatozoa was 55,65 ± 4,14 percent were first layer on inkubation 10 minutes, and the highestproportion Y spermatozoa was 62,19 ± 7,27 percent were fifth layer on inkubation 20 minutes treatment. The conclution from this study were : semen quality includes individual motility descending mean 29,25 percent from fresh semen and percentase of spermatozoa viability descending mean 38,44 percent. The highest individual motility there were on fifth layer sentrifugation treatment was 56,19 ± 1,90 percent, viability there were first layer on inkubation 20 minutes treatment was 59,65 ± 4,51 percent, and highest concentration there were on fifth layer sentrifugation treatment was 64,90 ± 24,16 million/ml. The highest spermatozoa X there were first layer on inkubation 10 minutes was 55,65 ± 4,14 percent, and the highest spermatozoa Y there were fifth layer on inkubation 20 minutes treatment was 62,19 ± 7,27 percent. The sugggestion from this study was to got spermatozoa X with the higher number is suggest to used first layer inkubation 10 minutes treatment and to got spermatozoa Y with the higher number is suggest to used fifth layer inkubation 20 minutes treatment. It was need to be done further research about effect of density and speed with different treatment of sentrifugation.
