Abstract
Stroberi di Indonesia, khususnya di daerah dataran tinggi telah dibudidaya secara komersial. Perbanyakan tanaman stroberi bisa dilakukan melalui biji, stolon atau kultur jaringan. Kultur jaringan, yaitu menggunakan jaringan meristem harus menggunakan bantuan ZPT. Oleh karena itu, perlu mencari konsentrasi ZPT yang optimal untuk mendukung perbanyakan eksplan yang dipropagasi. Tujuan dari penelitian ini adalah untuk mendapatkan konsentrasi perendaman Thidiazuron (TDZ) dan komposisi media dengan zat pengatur tumbuh yang tepat dalam menginduksi perbanyakan tunas tanaman stroberi dengan hipotesis, eksplan tanaman stroberi dengan pre-treatment Thidiazuron (TDZ) dan ZPT Thidiazuron (TDZ) sebagai media kultur dengan konsentrasi sekitar 0,5-1 ppm yang lebih banyak menginduksi tunas. Penelitian dilakukan di Lab. Kultur Jaringan BALITJESTRO, Batu, Malang, mulai Bulan September-Desember 2015. Alat yang digunakan meliputi: autoklaf, Laminar Air Flow (LAF), botol kultur, tabung Erlenmeyer, pipet, skala, penggaris, kertas label, gelas ukur, cawan petri, skapel (pen knife), timbangan analitik, hot plate, pH meter, pinset, dan kalkulator. Bahan yang digunakan adalah meristem batang stroberi varietas Early Brite serta ZPT yaitu TDZ, NAA, dan BAP. Rancangan yang digunakan adalah RAL faktorial perlakuan pre-treatment TDZ dan perlakuan media MS + ZPT. Perlakuan perendaman eksplan pada TDZ dengan menggunakn konsentrasi 0 ppm, 0,5 ppm, dan 1 ppm selama satu jam, perlakuan media dengan MS0, Media MS + TDZ 1 ppm, Media MS + BAP 0.5 ppm + NAA 0.025 ppm, dan Media MS + TDZ 1 ppm + BAP 0.5 ppm + NAA 0.025 ppm. Pengamatan dilakukan saat 3 HST sampai 56 HST. Pengamatan komponen pertumbuhan meliputi: persentase jumlah eksplan hidup perminggu, persentase jumlah eksplan membentuk tunas perminggu, waktu muncul tunas, waktu muncul akar, panjang eksplan, jumlah daun, jumlah akar, panjang akar, diameter tunas, dan jumlah tunas. Data yang diperoleh dianalisis dengan menggunakan uji F pada taraf 5% . Apabila terdapat pengaruh nyata, maka dilanjutkan dengan uji lanjut BNT pada taraf 5%. Hasil penelitian menunjukkan adanya interaksi antara pre-treatment TDZ dan perlakuan media MS + ZPT terhadap diameter tunas dan jumlah tunas. Perlakuan T2M3 dan T3M4 menunjukkan diameter tunas yang lebih besar daripada perlakuan lain. Jumlah tunas pada perlakuan T2M2 (19 tunas) lebih banyak dibandingkan dengan perlakuan lain, namun tidak berbeda nyata dengan perlakuan T1M4, T2M4, dan T3M2. Perlakuan perendaman eksplan pada zat pengatur tumbuh TDZ (pre-treatment), tidak berpengaruh nyata terhadap peubah yang diamati. Pada faktor media, perlakuan M3 menunjukkan hasil pembentukan planlet yang lebih baik dibandingkan perlakuan lain.
English Abstract
Strawberries in Indonesia, especially in the plateau area has been cultivated commercially. Propagation of strawberry plants can be done through seeds, stolon or tissue culture. Tissue culture, which is using meristem tissue should use the help of ZPT. Therefore, it is necessary to find the optimal concentration of ZPT to support propagation of explants which propagated. The purpose of this research is to obtain a concentration of soaking the explants in Thidiazuron (TDZ) and the composition of the media with growth regulators to induce the multiplication of shoots of strawberry plants, with the hypothesis explants of strawberry plants with pre-treatment Thidiazuron (TDZ) and PGR Thidiazuron (TDZ) as a culture medium with a concentration of about 0.5-1 ppm were induces much more shoots. The study was conducted at Tissue Culture Laboratory Balitjestro, Batu, Malang, from September-December 2015. The tools used include: autoclaving, Laminar Air Flow (LAF), the culture bottles, Erlenmeyer flask, pipettes, scales, rulers, paper label, beakers, petri dishes, skapel (pen knife), an analytical balance, hot plate, pH meter, tweezers, and a calculator. Materials used are meristem stem strawberry varieties of Early Brite and ZPT which is TDZ, NAA, and BAP. The design used was completely randomized design factorial, pre-treatment of TDZ treatment and treatment of MS + ZPT media. Soaking treatment at TDZ explants using a concentration of 0 ppm, 0.5 ppm and 1 ppm for one hour, the medias treatment is using MS0, Media MS + TDZ 1 ppm, Media MS + BAP + NAA 0.5 ppm 0.025 ppm, and Media MS + TDZ 1 ppm + BAP + NAA 0.5 ppm 0.025 ppm. Observations were carried out at three days after planting to 56 days after planting. Observation components of growth include: percentage of live explants per week, the percentage of explants forming shoots per week, time of emerging shoots, time appeared roots, explant length, number of leaves, number of roots, root length, shoot diameter, and number of shoots. The data were analyzed using the F test at 5% level. If there is a real effect, then followed by a further test LSD at 5% level. Result showed that there is interaction between pretreatment TDZ and treatment media MS + PGR towards shoot diameter and number of shoots. Treatment T2M3 and T3M4 indicates shoot diameter bigger than other treatments. The treatment of T2M2 (19 shoots) produce higher number of shoots than the other treatments, but not significantly different from treatment of T1M4, T2M4 and T3M2. Soaking treatment explants on plant growth regulator TDZ (pretreatment), did not significantly affect the observed variables. In the media factor, M3 treatments shows the results of plantlets forming better than the other treatment.
