Abstract

Tomat merupakan salah satu komoditas sayuran yang digemari oleh seluruh kalangan masyarakat. Hal tersebut dapat dilihat dari keunggulannya dalam memenuhi beberapa fungsi kehidupan seperti fungsi pemenuhan kebutuhan pangan, fungsi pemenuhan kebutuhan ekonomi, fungsi kesehatan, dan fungsi estetika. Namun, adanya serangan hama dan penyakit tanaman tomat menjadi faktor pembatas dalam produksi tomat. Salah satu penyakit tanaman tomat yang dapat menurunkan hasil yang sangat besar adalah penyakit antraknosa yang disebabkan oleh Colletotrichum sp. Pengendalian yang sering dilakukan ialah dengan pestisida sintetik yang memiliki dampak buruk bagi lingkungan. Oleh karena itu dibutuhkan pengendalian yang bersifat ramah lingkungan seperti penggunaan pestisida hayati. Saat ini agen hayati yang mulai banyak mendapat perhatian adalah khamir. Oleh karena itu penelitian ini dilakukan untuk memperoleh dan mengetahui potensi khamir yang terdapat pada buah tomat dan daun tanaman tomat sebagai agen antagonis patogen Colletotrichum sp. penyebab penyakit antraknosa pada tomat. Penelitian dilaksanakan di Laboratorium Mikologi, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya, Malang pada bulan Maret – September 2016. Isolasi khamir dilakukan pada daun, kulit buah, dan daging buah tomat yang sehat yang diperoleh dari lapang. Khamir yang diperoleh selanjutnya akan dilakukan uji antagonis terhadap patogen Colletotrichum sp. secara in-vitro dan in-vivo. Uji antagonis menggunakan RAL dengan 3 kali ulangan. Data yang diperoleh dari pengujian antagonis khamir terhadap patogen Colletotrichum sp. dianalisis menggunakan analisis ragam (Anova) dan apabila terdapat perbedaan yang nyata, akan dilanjutkan dengan menggunakan uji Beda Nyata Jujur (BNJ) pada taraf 5%. Khamir yang didapat dari isolasi kulit buah tomat, daging buah tomat, dan daun tanaman tomat adalah Ascoidea sp. (1)., Zygosaccharomyces sp., Pichia sp.(1), Cyniclomyces sp., Barnettozyma sp., Ambroxiozyma sp., Ascoidea sp. (2), Coccidiascus sp. (1), Coccidiascus sp. (2), Candida sp., Pichia sp. (2), Metschnikowia sp., Kodamaea sp., Saturnispora sp., Wickerhamiella sp., dan Kluyveromyces sp. 16 isolat khamir yang diujikan pada uji antagonis terhadap patogen Colletotrichum sp. secara in-vitro mampu menghambat pertumbuhan patogen Colletotrichum sp. dan menunjukan pengaruh yang berbeda nyata. Namun pada uji antagonis terhadap patogen Colletotrichum sp. secara in-vivo menunjukkan pengaruh yang tidak berbeda nyata. Khamir yang berpotensi paling baik dalam mengendalikan patogen Colletotrichum sp. ialah Ambroxiozyma sp. dengan daya hambat sebesar 30,87 %, masa inkubasi selama 6 hari, dan intensitas penyakit sebesar 9,34%.

English Abstract

Tomatoes are one of vegetables favored by all class of community. It can be seen from its superiority to fulfill many functions of life such as food needs function, the function of economic needs, health function, and aesthetic functions. However, the presence of pests and diseases of tomato plants become the limiting factor in the production of tomatoes. One of tomato plant diseases that can lower a very big result is anthracnose caused by Colletotrichum sp. Control is often done is to synthetic pesticides that have adverse environmental impacts. Therefore it takes control of the environmentally friendly such as the use of biological pesticides. Currently the biological agents that started a lot of attention was yeast. Therefore, this research was conducted to obtain and know the potential of yeast contained in the fruit of tomatoes and leaves of tomato plants as an antagonist agent pathogen Colletotrichum sp. the caused of anthracnose on tomatoes. The research was conducted at the Laboratory of Mycology, Department of Plant Pests and Diseases, Faculty of Agriculture, Brawijaya University on March until September 2016. Isolation of yeasts carried on the leaves, fruit peel and flesh of healthy tomatoes that obtained from the fields. The yeasts have been obtained will be tested antagonist against pathogens Colletotrichum sp. in ainvitro and in-vivo manner. This experiment used a completely randomized design (CRD) and each treatment were replicated 3 times. The data obtained from the antagonistic test were analyzed using analysis of variance (ANOVA) and if there is a real difference, will be continued by using test Honestly Significant Difference (HSD) at 5% level. Yeast derived from skin insulation tomatoes, tomato pulp, and leaves of tomato plants is Ascoidea sp. (1)., Zygosaccharomyces sp., Pichia sp. (1), Cyniclomyces sp., Barnettozyma sp., Ambroxiozyma sp., Ascoidea sp. (2), Coccidiascus sp. (1), Coccidiascus sp. (2), Candida sp., Pichia sp. (2), Metschnikowia sp., Kodamaea sp., Saturnispora sp., Wickerhamiella sp., and Kluyveromyces sp. 16 yeast isolates were tested on the antagonist test against pathogens Colletotrichum sp. in a invitro mannerto inhibit the growth of pathogenic Colletotrichum sp. and showed a significantly different effect. But in the antagonist test against pathogens Colletotrichum sp. in a in-vivo manner showed effect not significantly different. The most good potential yeast to control pathogens Colletotrichum sp. is Ambroxiozyma sp. by inhibition of 30,87%, an incubation period of 6 days, and the intensity of the disease by 9,34%.
