Abstract

Spodoptera litura merupakan hama yang bersifat polifag dan kosmopolit dengan kisaran inang yang luas (Su et al., 2012). Hama S. litura terbukti resisten terhadap berbagai insektisida kimia sehingga menyebabkan kegagalan panen (Ahmad et al., 2007), sehingga diperlukan alternatif pengendalian yang efektif dan ramah lingkungan, seperti penggunaan jamur Beauveria bassiana. Jamur B. bassiana merupakan jamur patogen serangga yang berpotensi untuk mengendalikan hama S. litura (Asi et al., 2013). Jamur B. bassiana mudah diperbanyak secara in vitro dengan media pertumbuhan buatan (Herlinda et al., 2006). Media sederhana, murah, namun tetap mengandung sumber nutrisi diperlukan dalam produksi massal jamur B. bassiana (Raimbault, 1998; Soccol dan Vandenberghe, 2003). Molase, air kelapa, dan tepung kulit udang dapat dimanfaatkan sebagai media pertumbuhan buatan jamur patogen serangga. Tujuan penelitian ini adalah menentukan perbedaan pengaruh media pertumbuhan pada kerapatan konidia, viabilitas, dan patogenisitas jamur B. bassiana terhadap hama S. litura. Penelitian ini dilaksanakan di Laboratorium Hama dan Penyakit Tumbuhan, Sub Lab. Nematologi dan Mikologi, Jurusan HPT FP UB pada bulan Februari sampai Mei 2016. Isolat jamur B. bassiana yang digunakan dalam penelitian ini diperoleh dari koleksi jurusan HPT FP UB. Perbanyakan isolat jamur B. bassiana menggunakan media SDAY.Penelitian ini menggunakan media pertumbuhan dari molase, air kelapa, dan tepung kulit udang dengan konsentrasi masing-masing 5% dan 10%. Jamur B. bassiana yang diperbanyak pada media molase, air kelapa, dan tepung kulit udang diinkubasi selama 15 hari untuk dilakukan pengamatan kerapatan, viabilitas, dan patogenisitas terhadap larva S. litura. Hasil penelitian antara lain kerapatan konidia dan viabilitas jamur B. bassiana diperbanyak pada media air kelapa 10% lebih tinggi daripada media lain. Mortalitas hama S. litura lebih tinggi pada perlakuan media tepung kulit udang 10% dibandingkan media lain. Rerata waktu kematian larva S. litura tersingkat akibat infeksi jamur B. bassiana yang diperbanyak pada media molase, air kelapa, dan tepung kulit udang antara 3,45-3,77 hari. Media air kelapa 10% dan media tepung kulit udang 10% dapat digunakan sebagai media alternatif yang efektif, murah, dan mudah diperoleh untuk perbanyakan jamur B. bassiana.

English Abstract

Spodoptera litura is a polyphagous and cosmopolitan pest with wide range of host (Su et al., 2012).S. litura reported resistant to many chemical insecticides (Ahmad et al., 2007), so it’s necessary to find effective and friendly alternative control, such as the use of Beauveria bassiana. B. bassiana is an entomopathogenic fungi that potential to control pest S. litura (Asi et al., 2013). B. bassiana is easy to propagated in vitro condition with artificial growth media (Herlinda et al., 2006). Simple and inexpensive media, but still contains source of nutrients required to the mass production of B. bassiana (Raimbault, 1998; Soccol and Vandenberghe, 2003). Molasses, coconut water, and shrimp shell powder can be used as artificial growth media of entomopathogenic fungus. The aim of this research was to determine the effect of different growth media on conidia density, vibility, and pathogenicity of B. bassiana against S. litura. This research was conducted at the Laboratory of Pest and Plant Disease, Sub Laboratory of Nematology and Micology, Department of Pest and Plant Disease, Faculty of Agriculture, University of Brawijaya, from February until May 2016. The isolate of B. bassiana that used in this research was obtained from Department of Pest and Plant Disease, University of Brawijaya. The propagation of B. bassiana using SDAY media. This research used artificial growth media from molasses, coconut water, and shrimp shell powder with each concentration 5% and 10%. B. bassiana that propagated in molasses, coconut water, and shrimp shell media were incubated in 15 days for observation of conidia density, viability, and pathogenicity against S. litura larvae. The results of this research were conidia density and viability of B. bassiana that propagated in 10% coconut water media was the highest of other media. Mortality of S. litura larvae with 10% shrimp shell powder media was the highest than other media. Mean time to death of S. litura infected by B. bassiana that propagated in molasses, coconut water, and shrimp shell powder media was range between 3.45 to 3.77 days. 10% coconut water media and 10% shrimp shell media can be used as effective and inexpensive artificial growth media for mass production of B. bassiana.
