Abstract

Ulat grayak (Spodoptera litura Fabricius) merupakan salah satu jenis hama penting yang menyerang tanaman palawija dan sayuran di Indonesia. S. litura bersifat polifag yang mempunyai kisaran inang yang luas. Jamur entomopatogen Beauveria bassiana berpotensi sebagai agens pengendali hayati berbagai spesies hama. Jamur B. bassiana mempunyai kapasitas reproduksi yang tinggi, mudah diproduksi dan pada kondisi yang kurang menguntungkan dapat membentuk spora. Meskipun B. bassiana menguntukan namun ada kekurangan. Virulensi B. bassiana beragam dan rendah apabila diaplikasi di lapang. Upaya untuk meningkatkan virulensi B. bassiana dengan kombinasi penambahan ekstrak larva inang di media PDA dan teknik inokulasi. Penelitian ini dilaksanakan di Laboratorium Nematologi dan Mikologi, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya, Malang mulai bulan April sampai September 2015. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) dengan delapan perlakuan dan tiga ulangan, sehingga terdapat 24 satuan percobaan. Perlakuan meliputi delapan kombinasi teknik inokulasi B. bassiana yang ditambah ekstrak larva pada media produksi PDA. Uji hayati menggunakan teknik inokulasi semprot daun, semprot larva, celup daun, dan celup larva dalam suspensi jamur B. bassiana pada median PDA dengan dan tanpa penambahan ekstrak larva. Setiap ulangan terdiri dari 20 larva S. litura instar kedua. Parameter pengamatan yang dilakukan yaitu viabilitas konidia, mortalitas S. litura, dan nilai LT50. Mortalitas S. litura tertinggi (71,67%) dihasilkan oleh infeksi jamur B. bassiana dari media PDA yang ditambahkan ekstrak larva dan larva dicelup dalam suspensi konidia. Mortalitas S. litura terendah (45,00%) dihasilkan oleh infeksi jamur B. bassiana dari media PDA tanpa penambahan ekstrak larva dan suspensi konidia disemprotkan ke daun sebagai pakan larva. Nilai LT50 S. litura tersingkat (4,58 hari) dihasilkan oleh infeksi jamur B. bassiana dari media PDA dan larva dicelup dalam suspensi konidia. Nilai LT50 S. litura terlama (8,09 hari) dihasilkan oleh infeksi jamur B. bassiana dari media PDA tanpa penambahan ekstrak larva dan suspensi konidia disemprotkan ke daun sebagai pakan larva.

English Abstract

Armyworm (Spodoptera litura Fabricius) is one of the important pests that attack crops and vegetables in Indonesia. S. litura are polifag and an important pest leaves because it has a wide host range. Entomopathogenic fungi Beauveria bassiana is potentially fungi in controlling various pest species. B. bassiana fungi has a high reproductive capacity, easily produced and at less favorable conditions can form spores. However, virulence of B. bassiana diverse and tend to be lower when applied in the field. Efforts to increase the virulence of B. bassiana by adding extracts larvae on media propagation and inoculation techniques. This research was conducted at the Laboratory Nematology and Mycology, Department of Pests and Plant Diseases, Faculty of Agriculture, University of Brawijaya, Malang from April to September 2015. These experiments using a completely randomized design with eight treatment and three replication, so there are 24 test unit. The treatment includes eight combination of inoculation technique B. bassiana plus extracts of larvae on media production. Bioassay was done by using application leaf spray, larvae spray, leaf dip, and larvae dip on the fungus B. bassiana suspension on the type of PDA media with and without extract larvae. Each treatment consist of 20 instar second instar larvae of S. litura. Parameter observations made are mortality S. litura and LT50 value. The highest S. litura mortality (71,67%) by fungal infection B. bassiana on PDA medium supplemented with larval extract and the larvae was dipped in a suspension of conidia. The lowest S. litura mortality (45,00%) by fungal infection B. bassiana on PDA medium supplemented without the addition of larval extracts and a suspension of conidial was sprayed onto the leaves as food for the larvae. The shortest LT50 value (4,58 days) by fungal infection B. bassiana on PDA medium supplemented with larval extract and the larvae was dipped in a suspension of conidia. The longest LT50 value (8,09 days) by fungal infection B. bassiana on PDA medium supplemented without the addition of larval extracts and a suspension of conidial was sprayed onto the leaves as food for the larvae.
