Abstract

Beauveria bassiana (Balsamo) Vuillemin memiliki potensi besar dalam mengendalikan hama utama pada berbagai komoditi tanaman. Jamur B. bassiana dapat digunakan dalam mengendalikan larva Spodoptera litura (Lepidoptera: Noctuidae). Larva S. litura termasuk hama penting pada tanaman kedelai. Aplikasi B. bassiana di filoplan tanaman kedelai telah banyak dilakukan, namun tingkat persistensi jamur ini belum diketahui. Sinar matahari dapat menghambat aktivitas konidia B. bassiana sehingga persistensi semakin singkat. Persistensi dan virulensi konidia B. bassiana terhadap larva S. litura di filoplan tanaman akibat pemaparan sinar matahari bergantung pada lama pemaparan. Semakin lama konidia B. bassiana di filoplan tanaman dipaparkan sinar matahari maka persistensi dan virulensi semakin rendah. Data persistensi dan virulensi merupakan langkah awal untuk menentukan frekuensi aplikasi B. bassiana di filoplan kedelai sebagai agens pengendali larva S. litura. Penelitian ini bertujuan untuk mengukur persistensi B. bassiana di filoplan kedelai setelah konidia dipaparkan sinar matahari selama 96 jam dan mengukur virulensi B. bassiana di filoplan kedelai terhadap larva S. litura setelah konidia dipaparkan sinar matahari selama 96 jam. Penelitian dilakukan di Laboratorium Nematologi dan Mikologi, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya pada bulan Februari 2015 sampai Juni 2015. Aplikasi pada filoplan kedelai dilakukan di Greenhouse III, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya. Penelitian ini menggunakan percobaan Rancangan Acak Lengkap (RAL). Perlakuan aplikasi yaitu pemaparan konidia B. bassiana di filoplan kedelai selama 0 (kontrol), 3, 6, 9, 12, 24, 48, 72, dan 96 jam. Masing-masing perlakuan diulang sebanyak 4 kali. Parameter pengamatan dalam penelitian ini antara lain viabilitas konidia B. bassiana, mortalitas larva S. litura, dan lama kematian. Data dari hasil penelitian dianalisis menggunakan uji ANOVA, apabila hasil menunjukkan pengaruh yang nyata maka dilanjutkan menggunakan uji Duncan pada taraf kepercayaan 95%. Pemaparan sinar matahari pada konidia B. bassiana di filoplan kedelai dengan lama pemaparan yang berbeda secara nyata berpengaruh terhadap persistensi dan virulensi konidia B. bassiana terhadap S. litura. Pemaparan sinar matahari selama 96 jam mampu menurunkan viabilitas konidia B. bassiana sebesar 81,72% dan waktu paruh 28,69 jam. Persentase mortalitas larva S. litura pada perlakuan pemaparan 0 jam mencapai 67,50% dan rerata waktu kematian 4,43 hari, sedangkan pada perlakuan pemaparan 96 jam menurun menjadi 6,25% dan rerata waktu kematian 6,75 hari.

English Abstract

Beauveria bassiana (Balsamo) Vuillemin has great potential in controlling major pests on a wide range of crop commodities. B. bassiana fungus can be used in control the larvae of Spodoptera litura (Lepidoptera: Noctuidae). The S. litura larvae was important pests of soybean. Application B. bassiana on soybean phylloplane have been studied, but the persistence level of the fungus is not known yet. Sunlight intensity can inhibit the activity of B. bassiana conidia so the persistence become shorter. The persistence and virulence of B. bassiana conidia on plants phylloplane due to sunlight exposure depends on the duration of exposure. The longer conidia of B. bassiana on plants phylloplane exposed to sunlight, the persistence and virulence become shorter. The persistence and virulence are the main step to establish the frequency application of B. bassiana on soybean phylloplane as control agent larvae of S. litura. This study aims to measure the persistence of B. bassiana conidia on soybean phylloplane after exposed to sunlight for 96 hours and measure the virulence of B. bassiana conidia on soybean phylloplane after exposed to sunlight for 96 hours against larvae of S. litura. The study was conducted in Nematologi and Mycology Laboratory, Department of Plant Pests and Diseases, Faculty of Agriculture, University of Brawijaya in February 2015 to June 2015. The application on soybean filoplan did in Green House III, Department of Plant Pests and Diseases, Faculty of Agriculture, University of Brawijaya. This research used experimental completely randomized design (CRD). The treatment of applications was exposure of B. bassiana conidia in soybean filoplan for 0 (control), 3, 6, 9, 12, 24, 48, 72, and 96 hours. Each treatment was repeated 4 times. Parameters of observation in this study include B. bassiana conidia viability, mortality larvae of S. litura long dead and death. Data from the study were analyzed using ANOVA test, if the results show a marked influence hence continued using Duncan test at 95% confidence level. Sunlight exposure on B. bassiana conidia on soybean phylloplane with different duration significantly affect the persistence and virulence of conidia of B. bassiana against larvae of S. litura. Sunlight exposure for 96 hours abled to reduce the viability of conidia of B. bassiana by 81,72% and the half-life time by 28,69 hours. The percentage mortality of larvae of S. litura on exposure treatment 0 hours reached 67,50% and the mean time of death was 4,43 days, while in the 96 hours exposure treatment decreased to 6,25% and the mean death time was 6,75 days.
