Abstract
Ulat grayak (Spodoptera litura F.) merupakan hama penting karena bersifat polifag sehingga berpotensi menjadi hama pada berbagai jenis tanaman. Pengendalian S. litura dapat memanfaatkan virus serangga SlNPV. Salah satu faktor yang berpengaruh terhadap penurunan persistensi dan infektifitas SlNPV adalah sinar ultraviolet. Penambahan bahan pembawa yang dicampurkan pada isolat SlNPV dapat mengurangi pengaruh negatif ultraviolet. Tujuan penelitian ini adalah mengetahui perbedaan persistensi dan infektivitas SlNPV yang ditambahkan kaolin dengan yang tidak ditambahkan kaolin dibawah pengaruh lama penyinaran ultraviolet yang berbeda. Penelitian dilaksanakan di Laboratorium Biologi, Jurusan Hama Penyakit Tumbuhan, Balitkabi, Kendalpayak, Kabupaten Malang. Pelaksanaan penelitian dimulai bulan April hingga Juni 2013. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) pola faktorial (2x4) dengan 3 ulangan. Parameter pengamatan pada larva S.litura meliputi stop feeding, mortalitas, metamorphosis larva menjadi pupa dan metamorphosis pupa menjadi imago. Hasil penelitian menunjukkan bahwa penambahan bahan pembawa berupa kaolin dapat mempertahankan persistensi SlNPV terhadap lama penyinaran ultraviolet. Persentase mortalitas larva S.litura pada perlakuan penambahan kaolin tanpa penyinaran sebesar 100%; perlakuan tanpa kaolin tanpa penyinaran sebesar 93,33%; perlakukan penambahan kaolin lama penyinaran 1 jam sebesar 83,33%; perlakukan tanpa kaolin lama penyinaran 1 jam sebesar 76,66%; perlakukan penambahan kaolin lama penyinaran sinar UV 3 jam sebesar 43,33%; perlakuan penambahan kaolin lama penyinaran sinar UV 4 jam sebesar 36,66%; perlakuan tanpa kaolin lama penyinaran sinar UV 3 jam sebesar 23,33%; dan perlakukan tanpa kaolin lama penyinaran sinar UV 4 jam sebesar 20,00%.
English Abstract
Spodoptera litura F. (Lepidoptera: Noctunidae) is an important poliphagous pest which means it could potentially become pests on different types of plants. The control of S. litura can be done by using insect virus SlNPV. One of the factors contributing to the reduction in persistence and infectivity of SlNPV is ultraviolet light. The addition of carrier on SlNPV isolates can reduce the negative influence of ultraviolet. The purpose of the research is to know the influence addition of kaolin and not kaolin towards SlNPV persistence and infectivity due to the effects of long UV rays irridiation. The research was carried out at Laboratorium Biologi, Jurusan Hama Penyakit Tumbuhan, Balitkabi, Kendalpayak, Regency of Malang. The research began on April to June 2013. The design of experiment used in this research was Completely Randomized Design (RAL) factorial pattern (2 x 4) with three replicates. Parameters of S. litura larvae observations including stop-feeding, mortality, larval metamorphosis into pupae stage and pupal metamorphosis into imago stage. The results showed that the addition of carrier in the form of kaolin can maintain persistence of SlNPV against long period of ultraviolet irradiation. The mortality percentage of larvae S. litura with kaolin application and no UV rays irradiation is 100%; without kaolin and no UV rays irradiation is 93,33%; with kaolin application and 1-hour long UV rays irradiation is 83,33%; without kaolin on 1-hour long UV rays irradiation is 76,66%; kaolin application and 3 hour long UV rays irradiation is 43,33%; with kaolin application on 4 hour long UV rays irradiation is 36,66%; without kaolin and 3 hour long UV rays irradiation is 23,33%; and without kaolin on 4 hour long UV rays irradiation is 20.00%.
