Abstract

Entomopatogenic Nematode (EPN) adalah salah satu agen biologis dengan kemampuan untuk mengontrol hama tanaman. Dua genus EPN dengan peran penting agen biologis adalah genus steinernema dan genus heterorhabditis. Kemampuan EPN mendistribusikan, mempertahankan, menemukan tuan rumah, dan mereproduksi di bidang dipengaruhi oleh jenis tanah, kelembaban, suhu dan tuan rumah. Untuk mendapatkan EPN, isolat lokal diperlukan untuk kegiatan eksplorasi, yang kemudian dilanjutkan dengan isolasi dan identifikasi. Tujuan dari penelitian ini adalah untuk memahami genus EPN dan kepadatan populasinya di ladang jagung, kedelai dan kubis dan untuk mengakui virulasinya terhadap larva spodoptera Litura. EPN diperoleh melalui eksplorasi di ladang jagung di Desa Ngijo, di bidang kedelai di bidang penelitian Kantor Penelitian Kacang dan umbi di Kendalpayak Vilage, dan di lapangan kubis di Desa Bumiaji. EPN diidentifikasi berdasarkan gejala pewarnaan pada kutikula dan morfologi. Selanjutnya, EPN diperiksa untuk laju virulensi terhadap larva S. Litura. Hasil isolat EPN menunjukkan bahwa populasi tertinggi ditemukan di bidang kubis untuk 23.264 EPN / 0,25 ml, diikuti di bidang jagung untuk 16.976 EPN / 0,25 ml, dan terendah di bidang kedelai untuk 15.664 EPN / 0,25 ml. Hasil identifikasi EPN di bidang kubis menunjukkan genus EPN Steinernema dan genus heterorhabditis. Di ladang kedelai dan jagung, Genus EPN Steinernema jelas. Hasil uji virulensi EPN terhadap Larva S. Litura menunjukkan bahwa tiga isolat EPN memiliki tingkat virulensi serupa dalam mengontrol larva.

English Abstract

Entomopathogenic nematode (EPN) is one of biological agents with capability to control crop pest. Two genus of EPN with significant role of biological agents are Genus Steinernema and Genus Heterorhabditis. The capability of EPN of distributing, defending, finding host, and reproducing in the field is affected by soil type, humidity, temperature and host. To obtain EPN, the local isolate is necessary for the exploration activity, which is then continued by isolation and identification. The objective of research is to understand the EPN genus and its population density in the corn, soybean and cabbage fields and to acknowledge its virulence against larvae Spodoptera litura. EPN is obtained through exploration in the corn field at Ngijo Village, in the soybean field at the research field of Beans and Tubers Research Office at Kendalpayak Vilage, and in the cabbage field at Bumiaji Village. EPN is identified based on the coloration symptom on the cuticle and morphology. Subsequently, EPN is examined for its virulence rate against larvae S. litura. Result of EPN isolate indicates that the highest population is found in the cabbage field for 23,264 EPN/0.25 ml, followed in the corn field for 16,976 EPN/0.25 ml, and the lowest in the soybean field for 15,664 EPN/0.25 ml. Result of EPN identification in the cabbage field is showing the EPN Genus Steinernema and Genus Heterorhabditis. In soybean and corn fields, EPN Genus Steinernema is obvious. Result of virulence test of EPN against larva S. litura indicates that three isolates of EPN have similar virulence rate in control the larvae.
