Abstract

Sepsis adalah sindrom klinis yang disebabkan respon inflamasi terhadap infeksi. Sepsis induksi lipopolisakarida (LPS) menginisiasi inflamasi dan menyebabkan peningkatan produksi IL-1β sebagai indikator respon hospes terhadap patogen, serta penyebab kerusakan jaringan hepar. Tindakan pencegahan sepsis dapat dilakukan dengan pemberian daun sambiloto yang mengandung andrographolide sebagai antiinflamasi dan hepatoprotektor. Tujuan penelitian ini untuk mengetahui efek preventif ekstrak daun sambiloto berdasarkan kadar IL-1β dengan metode ELISA dan gambaran histopatologi hepar dengan pewarnaan haematoxylin-eosin (HE). Penelitian ini bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 20 hewan coba tikus putih jantan umur 8-12 minggu dan berat badan 150-200 gram, dibagi menjadi lima kelompok yaitu kontrol negatif, kontrol positif, P1 (250 mg/ kg BB), P2 (500 mg/ kg BB), dan P3 (1000 mg/ kg BB). Data kadar IL-1β dianalisis secara statistik menggunakan metode One-Way ANOVA dan uji BNJ (α=0,05). Hasil penelitian menunjukkan rata – rata kadar IL-1β serum berbeda nyata (p<0.05) antar kelompok. Kadar IL-1β P3 (512.33 ± 199.44) menunjukkan hasil yang terbaik dan gambaran histopatologi hepar yang tetap normal. Kesimpulan penelitian ini ekstrak daun sambiloto dosis 1000 mg/kg BB dapat dijadikan preventif untuk sepsis.

English Abstract

Septic is a clinical syndrome caused by an inflammatory response to infection. Septic lipopolysaccharide (LPS) induction initiates inflammation and leads to increased IL-1β production as an indicator of host response to pathogens, also caused of hepatic tissue damage. Septic could prevented by sambiloto leaf administration that containing andrographolide as anti-inflammatory and hepatoprotector. The purpose of this research is to know the preventive effect of sambiloto leaf extract based on IL-1β level with ELISA method and liver histopathology with Haematoxylin-Eosin (HE) stain. This study is an experimental using Completely Randomized Design (CRD) with 20 male rats, 8-12 weeks old and 150-200 gram body weight, divided into five groups: negative control, positive control, P1 (250 mg / kg BW), P2 (500 mg / kg BW) and P3 (1000 mg / kg BW). Data of IL-1β levels were statistically analyzed using One-Way ANOVA method and BNJ test (α = 0.05). The results showed that mean IL-1β levels were significantly different (p<0.05) between the groups. Level of IL-1β P3 (512.33 ± 199.44) showed the best results and normal liver histopathology. The conclusion of this research is sambiloto leaf extract 1000 mg / kg BW dose can be used as preventive for septic.
