Abstract
Timbal (Pb) merupakan bahan kimia yang termasuk dalam kelompok logam berat dan berpotensi menjadi bahan toksik. Paparan Plumbum dapat melalui saluran pernafasan, pencernaan, dan permukaan kulit. Akumulasi Plumbum dalam tubuh dapat meningkatkan jumlah Reactive Oxygen Species (ROS) yang berdampak pada kerusakan organ, khususnya organ ginjal. Kerusakan ginjal dapat menyebabkan peningkatan Blood Urea Nitrogen (BUN). Madu Hutan Sumbawa merupakan salah satu madu yang memiliki kandungan flavonoid yang cukup tinggi, sehingga dapat digunakan sebagai antioksidan yang dapat menetralisir radikal bebas. Penelitian ini berfungsi untuk mengetahui pengaruh terapi preventif Madu Hutan Sumbawa terhadap kadar Blood Urea Nitrogen (BUN) dan gambaran histopatologi ginjal tikus putih (Rattus norvegicus) yang diinduksi Plumbum Asetat. Penelitian ini menggunakan 20 ekor tikus yang dibagi menjadi 5 kelompok perlakuan, yaitu kontrol negatif tanpa diberikan perlakuan, kontrol positif dengan pemberian Pb asetat 10 mg/ekor/hari selama 14 hari, dan kelompok terapi preventif dengan pemberian Madu Hutan Sumbawa 25 mg/kg BB, 50 mg/kg BB, 75 mg/kg BB selama 28 hari dan pemberian Pb Asetat dosis 10 mg/ekor/hari selama 14 hari pada hari ke-15 sampai hari ke-28. Setiap kelompok perlakuan terdiri dari 4 tikus. Pengukuran kadar BUN menggunakan metode spektrofotometri dan histologi ginjal menggunakan pewarnaan HE yang diamati dengan mikroskop cahaya. Kadar BUN dianalisa menggunakan analisa ragam ANOVA dan dilanjutkan uji Tukey (ɑ = 5%). Hasil penelitian menunjukkan pemberian terapi preventif madu hutan Sumbawa dapat menurunkan nilai kadar BUN dan memperbaiki kerusakan organ ginjal dengan dosis terbaik yaitu 75 mg/kg BB. Perbaikan gambaran histopatologi ginjal terlihat dengan tidak terjadinya pelebaran ruang bowman, piknosis pada glomerulus, lisis glomerulus, edema interstisial.
English Abstract
Plumbum’s exposure can be through inhalation, digestion and skin surface. Accumulation of plumbum in the body can increase the number of Reactive Oxygen Species (ROS) which can cause organ damage, especially kidney. Kidney’s damage can cause the increasing of Blood Urea Nitrogen (BUN). Sumbawa forest honey is one kind of honey which has high flavonoid content, therefore it can be used as an antioxidant that can neutralize the radicals. The aim of this research was to identify the effect of preventive therapy of sumbawa forest honey against levels of Blood Urea Nitrogen (BUN) and histopathological features of rat (rattus norvegicus) kidney that induced by plumbum acetate. This research used 20 rats, those were divided into 5 treatment groups, which were negative control group without treatment, positive control group which induced Pb acetate 10mg/mouse/day for 14 days, and preventive therapy group which administrate the sumbawa forest honey with 25mg/kg BW, 50mg/kg BW, 75 mg/kg BW doses for 28 days and also Pb acetate with 10mg/rat/days for 14 days on day 15th until 28th. Each treatment group consists of 4 rats. Levels of BUN was measured using spectrophotometry and kidney histological features was stained using HE stain which then observed using microscope. BUN levels were analyzed using ANOVA and continued with Tukey test (a=5%). The results showed that the administration of Sumbawa forest honey as preventive therapy can reduce levels of BUN content and improve the damage of kidney organ with the best dose of 75 mg/kg BW. Improvements in renal histopatologic features are seen in the absence of widening bowman spaces, pyknosis in the glomerulus, glomerular lysis and interstitial edema.
