Abstract
Asma merupakan penyakit kronik saluran pernafasan yang banyak dijumpai pada hewan dan manusia. Gejala asma yang mucul dapat diperparah oleh infeksi rongga mulut akibat paparan Lipopolisakarida (LPS) dari bakteri Gram negatif. Penelitian ini menggunakan ekstrak daun putri malu (Mimosa pudica) untuk terapi pada hewan model tikus (Rattus norvegicus) model asma. Kandungan flavonoid pada putri malu berperan sebagai antioksidan alami yang dapat menghambat radikal bebas. Tujuan penelitian ini mengetahui ekspresi iNOS dan mengetahui gambaran infiltrasi sel inflamatori pada jaringan bronkiolus pada hewan tikus model asma setelah diberi terapi ekstrak daun putri malu (Mimosa pudica L.). Penelitian ini menggunakan empat kelompok tikus, yaitu kelompok kontrol(A), kelompok asma (B), kelompok asma yang mendapat terapi daun putri malu dosis 500 mg/KgBB (C) dan dosis 1000 mg/KgBB (D). Tikus model asma disiapkan dengan cara pemberian sensitasi alergi dengan Ovalbumin (OVA) dengan dosis 10 μg/ml/ekor secara intraperitonial dan inhalasi serta pemberian LPS dari Phorphyromonas gingivalis dengan dosis 1 μg/ml/ekor secara intrasulkuler. Ekspresi iNOS dengan immunohistokimia dan infiltrasi sel inflamatori bronkiolus dengan pewarnaan HE diamati secara mikroskopis. Hasil penelitian menunjukkan pemberian terapi daun putri malu dengan dosis 500 mg/Kg BB dan 1000 mg/Kg BB mampu menurunkan ekspresi iNOS dan infiltrasi sel inflamatori bronkiolus pada tikus model asma secara signifikan (p<0,05). Ekstrak daun putri malu dosis 1000 mg/kg BB memberikan hasil terbaik terhadap penurunan ekspresi iNOS sebesar 73,63% dan penurunan infiltrasi sel inflamatori sebesar 79,53%. Kesimpulan dari penelitian ini adalah ekstrak daun Putri malu dapat digunakan sebagai terapi herbal pada tikus model asma.
English Abstract
Asthma is a chronic respiratory disease that often found in animals and humans. Asthma symptoms become more severe by oral infections due to exposure of Gram-negative bacterial lipopolysaccharide (LPS). The Mimosa pudica leaf extract were used as therapy agent on animal model of asthma rats (Rattus norvegicus). Flavonoid substance in Mimosa pudica leaf extract play a role to be nature antioxidant which has an activity to block ROS (Reactive Oxygen Species). The purpose of this research was to study the potency of Mimosa pudica leaf extract toward the expression of iNOS and bronchioles infiltration inflammatory cells on asthma rats. Four groups of rats (Rattus norvegicus) were used in this research control group, asthma group, and two groups with therapy of Mimosa pudica extract dose of 500 mg/Kg BW and 1000 mg/Kg BW. Asthma rats were prepared by sensitization of allergent conducted by intraperitonial injected and nebulized of 10μg/ml Ovalbumin (Ova) also intrasulcular injection of 1μg/ml Lipopolysaccharide from Phorphyromonas gingivalis. The expression of iNOS and bronchioles infiltration inflammatory cells histopathology were observed microscopically. The result showed that Mimosa pudica leaf extract therapy with both dose of 500 mg/Kg BW and 1000 mg/Kg BW decreasing iNOS expression and bronchioles infiltration inflammatory cells histopathology significantly (P<0.05). The use of1000 mg/kg BW dose Mimosa pudica leaf extract were the best theraphy to decrease expression of iNOS to be 73.63% and bronchioles infiltration inflammatory cells histopathology to be 79.53%. It can be concluded that Mimosa pudica leaf extract have possibility as herbal theraphy on asthma rats models.
