Abstract

Hiperkolesterolemia merupakan gangguan metabolisme yang disebabkan tingginya kolesterol didalam darah. Peningkatan metabolisme lipid menyebabkan timbulnya peroksidasi lipid yang mengakibatkan kerusakan sel, sehingga dibutuhkan antioksidan pada flavonoid ekstrak air daun sukun (Artocarpus altilis) yang berfungsi membantu sintesis kolesterol berlebih di dalam tubuh. Tujuan penelitian ini adalah mengetahui pengaruh terapi ekstrak air daun sukun terhadap penurunan kadar MDA dan perbaikan gambaran histopatologi tunika mukosa jejunum tikus hiperkolesterolemia. Penelitian ini menggunakan hewan model tikus dengan 5 kelompok perlakuan yaitu kontrol, hiperkolesterolemia, terapi ekstrak air daun sukun dosis 500 mg/kg BB, 1000 mg/kg BB dan 2000 mg/kg BB. Pengukuran kadar MDA menggunakan metode uji Thioarbituric Acid Reactive Substance (TBARC), dianalisa secara kuantitatif menggunakan one way ANOVA dan uji lanjutan tukey. Pengamatan histopatologi dilakukan dengan pewarnaan Hematoksilin Eosin (HE) diamati menggunaan miskroskop dan dianalisa secara kualitatif deskripstif. Hasil penelitian menunjukan bahwa pemberian terapi ekstrak air daun sukun dengan dosis 500 mg/kg BB, 1000 mg/kg BB dan 2000 mg/kg BB menurunan kadar MDA organ jejunum tikus hiperkolesterolemia secara signifikan (p<0,05). Dosis terapi 2000 mg/kg BB menurunkan kadar MDA sebesar 43,57% dan menunjukan perbaikan gambaran histopatologi tunika mukosa jejunum tikus hiperkolesterolemia. Kesimpulan penelitian ini yaitu ekstrak air daun sukun dapat menurunkan kadar MDA dan memperbaiki gambaran histopatologi tunika mukosa jejunum tikus hiperkolesterolemia.

English Abstract

Hypercholesterolemia is a metabolic disorder due to high cholesterol in the blood. Increasing of lipid metabolism causes damaged cell by high lipid peroxidase. Breadfruit leaf extract as antioxidant is needed to reduce high cholesterol level in the blood. The purpose of this research was to know the effect of breadfruit leaf extract therapy toward MDA level and jejunum histopathology on hypercholesterolemia. This research used rats which were divided into five groups: control, hypercholesterolemia, hypercholesterolemia treated with breadfruit leaf extract dosage of 500 mg/Kg BW, 1000 mg/Kg BW and 2000 mg/Kg BW group. Malondyaldehide (MDA) level was measured by Thioarbituric Acid Reactive Substances (TBARC) test and analyzed quantitatively by oneway ANOVA and continued by tukey test. Jejunum histopathology was staining by Hematoksilin-eosin (HE) and analyzed qualitatively. The result showed that breadfruit leaf extract therapy dosage of 500 mg/kg BW, 1000 mg/kg BW and 2000 mg/kg BW decreased MDA level of jejunum hypercholesterolemia rats significantly (p<0,05). The dose 2000 mg/kg BW therapy is the best dose that decreasing MDA level of 43,57% and repairing of jejunum histopathological appearance noticed by cell proliferation in tunica mucosa. The conclusion of this research was breadfruit leaf extract reduce MDA level and repaire the jejunum histopathology of tunica mucosa on hypercholesterolemia rats.
