Abstract

Pola makan dengan diet tinggi lemak dapat menyebabkan hiperkolesterolemia, yaitu suatu keadaan dimana kadar kolesterol dalam darah melebihi batas normal. Angka kejadian hiperkolesterolemia cukup tinggi dan dapat menyebabkan penyakit jantung koroner. Yoghurt susu kambing mengandung bakteri asam laktat, biopeptida, antioksidan, dan vitamin yang dapat menurunkan kadar kolesterol dalam darah. Tujuan penelitian ini adalah untuk mengetahui pengaruh terapi yoghurt susu kambing terhadap kadar Malondialdehida (MDA) dan ekspresi Tumor Necrosis Factor-α (TNF-α) pada organ hepar tikus ( Rattus norvegicus ) hiperkolesterolemia. Penelitian ini menggunakan 20 ekor tikus umur 10-12 minggu yang dibagi dalam 5 kelompok yaitu kelompok kontrol, hiperkolesterolemia, hiperkolesterolemia dan terapi dosis 300, 600, dan 900 mg/kg BB. Pembuatan hewan model hiperkolesterolemia dengan pemberian kuning telur puyuh rebus, minyak babi, dan asam kholat selama 14 hari dan terapi yoghurt susu kambing diberikan selama 28 hari. Kadar MDA diamati dengan metode Thiobarbituric Acid Reactivity Test ( TBA -RT) dan ekspresi TNF-α dengan metode imunohistokimia dengan menggunakan software Axio Vision . Data yang diperoleh dianalisis menggunakan SPSS versi 16.0 dengan analisis ragam One Way Analisis Of Variance (ANOVA) dan dilakukan analisis lebih lanjut dengan uji Tukey (p<0,01). Hasil One Way ANOVA kadar MDA dan ekspresi TNF- α pada tikus perlakuan menunjukkan adanya perbedaan yang sangat nyata (p<0,01). Terapi yoghurt susu kambing dosis 300 mg/kg BB dan 600 mg/kg BB mempunyai potensi menurunkan kadar MDA dan ekspresi TNF- α organ hati hewan model tikus hiperkolesterolemia. English Abstract

The habitual of consuming fatty foods triggers hypercholesterolemia which is characterized by increasing of cholesterol level in the blood higher than the limit. The prevalence of hypercholesterolemia is enormous and possible cause of death. The aim of this research was to study therapeutic effect of goat milk yogurt on the Malondialdehyde (MDA) levels and TNF-α expression in hypercholesterolemic rat’s liver. This research use d male rats, age of 10-12 weeks which was divided into five groups; control, hypercholestrolemia, hypercholesterolemia with goat milk yogurt therapy dose of 300, 600, and 900 mg/kg BW. The hypercholesterolemic rats was prepared by administered the egg yolk of boiled quail, lard, and cholic acid for 14 days. The goat milk yogurt therapy was conducted for 28 days by force feeding method. MDA level was measured using the Thiobarbituric Acid Reactivity Test ( TBA -RT) methode and TNF-α expression was observed using immunohistochemistry. Data was analysed by SPSS 16.0 version using one way Analysis of Variance (ANOVA), continued by Tukey test (p<0.01). The result of One Way ANOVA showed that the average of MDA level and TNF- α expression from each group was high significantly different (p<0.01). The goat milk yogurt therapy dose of 300 mg/kg BW and 600 mg/kg BW ha d a potensial to decrease MDA level and TNF- α expression in liver of hypercholesterolemic animal mode l. The goat milk yogurt therapy dose of 900 mg/kg BW is the most effective dose in decreasing MDA level ( 58.81% ) and TNF- α expression ( 83.88% ) in liver of hypercholesterolemic animal model.
