Abstract

Kitosan telah diketahui dapat berperan dalam mencegah terjadinya proses demineralisasi pada enamel gigi. Pada lingkungan asam, gugus amino pada kitosan dapat menangkap ion hidrogen yang menjadikannya bermuatan positif dan bersifat bioadhesif terhadap permukaan enamel, sehingga dapat berperan sebagai barier terhadap penetrasi asam. Dengan ukuran kecil yang dimilikinya, nanopartikel kitosan dapat menembus membran yang tidak dapat dilewati partikel yang lebih besar. Penelitian ini bertujuan untuk mengetahui perbedaan pengaruh pemberian pasta nanopartikel dan mikropartikel kitosan cangkang udang terhadap ketahanan kekerasan mikro permukaan enamel gigi. Penelitian ini menggunakan Vickers Hardness Tester untuk menghitung kekerasan mikro permukaan enamel. 27 gigi Premolar 1 RA yang dibagi ke dalam 3 kelompok, yaitu kelompok kontrol (-), kelompok perlakuan nanopartikel kitosan, dan kelompok perlakuan mikropartikel kitosan. Semua kelompok melewati proses demineralisasi selama 1 jam yang dilakukan 3x sehari selama 5 hari. Pada kelompok perlakuan, sebelum sampel melewati satu siklus proses demineralisasi, sampel diaplikasikan dengan pasta kitosan selama 5 menit. Hasil rerata selisih kekerasan mikro permukaan enamel sebelum dan sesudah perlakuan pada kelompok nanopartikel kitosan 106,11 HV dan pada kelompok mikropartikel kitosan 140,54 HV. Hasil uji independent T-test menunjukkan bahwa terdapat perbedaan yang signifikan antara kelompok nanopartikel kitosan dan mikropartikel kitosan pada sampel 2 dan 4, sedangkan pada sampel lainnya tidak terdapat perbedaan yang signifikan. Kesimpulannya adalah nanopartikel kitosan dapat mempertahankan kekerasan mikro permukaan enamel lebih baik dibandingkan dengan mikropartikel kitosan.

English Abstract

Chitosan has important roles in preventing enamel demineralization. In acid environment, chitosan’s amino group captures hydrogen ions, so it has positive charge that gives bioadhesive ability to negatively charged surfaces such as tooth enamel. Chitosan will be absorbed into enamel surface and form a layer that act as a barrier against acid penetration. The nanoparticle’s small size makes it possible for it to pass through the membrane that is impermeable to bigger particles. The aim of this study was to evaluate the difference effect of nanoparticle and microparticle chitosan from shrimp shell (Pandalus borealis) treatment towards the enamel microhardness endurance of enamel surface. 27 human maxillary 1st Premolar were divided into three groups; the negative control group, nanoparticles chitosan group, and microparticles chitosan group. All groups were through demineralization process for 1 hour, 3x per day during 5 days. Tooth samples on the treatment groups were pretreated with chitosan paste for 5 minutes before exposure to each cycle of demineralization. The microhardness of the enamel was measured using Vickers Hardness Tester, then the difference between microhardness before and after treatment was measured to evaluate the mineral loss. The mean difference microhardness result of nanoparticles chitosan group was 106,11 HV and microparticles chitosan group was 140,54 HV. Independent T-test result showed that there were statistical differences between nanoparticle and microparticle chitosan groups on 2nd and 4th sample, while other samples, there were no statistical differences between them. Nanoparticle chitosan could maintain the microhardness of enamel better than microparticle chitosan.
