Pengaruh Ekstrak Kulit Tomat (Solanum Lycopersicum) Terhadap Kadar Serum Kolesterol Total Pada Tikus Model Diabetes Melitus Tipe 2

Abstract

Diabetes Melitus (DM) merupakan kelainan metabolik yang ditandai dengan hiperglikemia yang terjadi karena kelainan sekresi insulin, kerja insulin, atau kedua-duanya. Kondisi hiperglikemia pada DM tipe 2 menyebabkan terjadinya peningkatan radikal bebas atau stres oksidatif yang dapat memicu kerusakan hepatosit. Hal ini dapat mempengaruhi penurunan protein plasma sehingga terjadi peningkatan kadar kolesterol darah. Oleh karena itu, dibutuhkan suatu antioksidan yang dapat menetralkan radikal bebas pada DM tipe 2 yang bertujuan untuk menghambat peningkatan kadar kolesterol darah. Tomat (Solanum lycopersicum) pada bagian kulit mengandung antioksidan yang lebih tinggi dari pada bagian daging buah seperti likopen, α-karoten, β-karoten, kuersetin, vitamin larut air dan larut lemak seperti vitamin A, vitamin E, vitamin C dan lutein. Kandungan ini dapat menghambat proses aktifitas radikal bebas sehingga proses oksidatif pada jaringan terhambat. Penelitian ini bertujuan untuk membuktikan pengaruh ekstrak kulit tomat dalam menurunkan kadar kolesterol total. Penelitian eksperimental ini menggunakan tikus putih Rattus norvegicus sebanyak 20 sample dipilih berdasarkan Simple Random Sampling dan dibagi menjadi lima kelompok yaitu KN (kontrol negatif), KP (kontrol positif model DM tipe 2 ), dan KP1, KP2, KP3 (model DM 2 diberi ekstrak kulit tomat dosis 50, 100, 150 mg/kgBB) selama 3 minggu kadar kolesterol total diukur dengan spektrofotometri. Data diuji menggunakan One Way Anova dengan signifikansi < 0,05. Hasil penelitian terdapat peningkatan kadar kolesterol total pada kelompok model DM tipe 2 dibandingkan dengan kontrol negatif, namun tidak signifikan, serta terdapat penurunan kadar kolesterol total pada pemberian dosis 150 mg/kgBB, namun tidak signifikan. Kesimpulan penelitian adalah pemberian ekstrak kulit tomat dengan dosis 50, 100, 150 mg/kgBB belum dapat menurunkan kadar kolesterol total pada tikus model DM tipe 2.

English Abstract

Diabetes mellitus (DM) is a metabolic disorder characterized by hyperglycemia resulting from defects in insulin secretion, insulin action, or both insulin secretion and insulin action. The condition of hyperglycemia in DM type 2 can be one of the factors that can increase free radical or oxidative stress that can lead to damage of hepatocytes. This can affect a decrease in plasma proteins resulting in increased blood cholesterol levels. Therefore, it takes an antioxidant that can neutralize free radicals in DM type 2 aims to prevent elevated blood cholesterol levels. Tomato skin (Solanum lycopersicum) contains antioxidants that are higher than flesh of tomato like lycopene, α-carotene, β-carotene, quercetin, water and fat soluble vitamin like vitamin A, vitamin E, vitamin C and lutein. This substance can inhibit the activity of free radicals so the process of oxidative in the tissue is inhibited. This study aims to prove the effect of tomato skinl extracts in lowering total cholesterol levels. This experimental research used white rats Rattus norvegicus as many as 20 samples selected by simple random sampling and divided into five groups, namely KN (negative control), KP (positive control models of DM type 2), and KP1, KP2, KP3 (model DM type 2 that given tomato skin extract doses of 50, 100, 150mg/kgBW) for 3 weeks total cholesterol levels were measured by spectrophotometry. Data were tested use One Way ANOVA with significance <0.05. The result of the research is there is an increase of total cholesterol levels in DM type 2 group compared to the negative control group, but not significant, and there is a decrease in total cholesterol levels in group that given 150 mg/kgBW of tomato skin extract, but not significant. Conclusion of this research is tomato skin extract with 50, 100, 150 mg/kgBW dose has not been able to reduce total cholesterol in white rats of DM type 2 models.
