Abstract

Diabetes mellitus (DM) merupakan suatu penyakit metabolik kompleks dengan kriteria diagnosis kadar plasma glukosa puasa ≥ 126 mg/dL atau kadar plasma glukosa acak atau toleransi glukosa 2 jam ≥ 200 mg/dL. Terdapat dua tipe utama DM yaitu tipe 1 dan 2 dimana DM tipe 1 disebabkan oleh defisiensi insulin absolut. DM berada di posisi 4-5 sebagai penyebab kematian di negara maju dan menjadi epidemi di negara berkembang. DM tipe 1 dapat menimbulkan komplikasi jangka panjang, salah satunya adalah nefropati. Penelitian ini bertujuan untuk membuktikan efektivitas pemberian air kelapa muda secara oral terhadap penurunan kadar ureum tikus yang diinduksi aloksan dengan melakukan studi eksperimental murni (true experimental design) menggunakan rancangan Randomized Post Test Only Controlled Group Design pada tikus Wistar jantan model DM Tipe 1. Sampel sebanyak 27 ekor tikus (n=9 tiap kelompok) dipilih berdasarkan Simple Random Sampling dan dibagi merata terhadap tiga kelompok yaitu kontrol negatif, kelompok kontrol positif dan kelompok perlakuan yang diberi air kelapa muda 4 mL/hari selama 14 hari yang diukur menggunakan spektrofotometri. Kemudian perolehan data kadar ureum diuji menggunakan statistik One Way ANOVA dengan taraf signifikansi 0,05. Hasil penelitian menunjukkan bahwa terdapat perbedaan bermakna kadar ureum antara dua kelompok perlakuan, yaitu kelompok kontrol negatif dan kelompok kontrol positif (p = 0,000), kelompok kontrol negatif dan kelompok perlakuan (p = 0,000), serta kelompok kontrol positif dan kelompok perlakuan (p = 0,000). Kesimpulan dari penelitian ini adalah pemberian air kelapa muda secara oral dapat menurunkan kadar ureum tikus diabetes yang diinduksi aloksan.

English Abstract

Diabetes mellitus represents one of complex metabolic disease with diagnostic criteria fasting plasma glucose level ≥ 126 mg/dL or random plasma glucose or 2-hour plasma glucose level ≥ 200 mg/dL. There are 2 major types of Diabetes : type 1 and 2 which type 1 DM is caused by absolute insulin deficiency. DM is in 4-5th position to cause mortality in advanced country and become epidemic in developing country. Type 1 DM can cause long term complications, one of them is nephropathy. This research was aimed to prove the effectivity of oral administration coconut water to reduce the level of urea on alloxan-induced rats by doing true experimental study using Randomized Post Test Only Controlled Group Design was conducted onto male Wistar strain rats Type 1 DM model. The total samples were 27 rats (n=9 for each group) and selected by simple random sampling and divided into three groups, they were negative control group, positive control group and treatment group that was given 4 mL/day coconut water for 14 days. The level of rats’ urea serum was measured by spectrophotometry. Result of this research was tested by statistic One Way ANOVA with significance degree 0,05. There were significant differences of urea serum level between two treatment groups, which were negative control group and positive control group (p = 0,000), negative control group and treatment group (p = 0,000), and positive control group with treatment group (p = 0,000). The conclusion is oral administration of coconut water can reduce urea serum level of alloxan-induced diabetes rats.
