Abstract

Penggunaan antibiotik banyak menimbulkan masalah seperti menimbulkan residu dan membuat bakteri menjadi resisten. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak kasar brotowali (T. cordifolia) terhadap bakteri V. harveyi secara In Vitro. Metode yang digunakan dalam penelitian ini adalah metode eksperimen. Rancangan percobaan yang digunakan adalah Rancangan Percobaan Acak Lengkap (RAL) dengan menggunakan 5 perlakuan ekstrak kasar brotowali yaitu konsentrasi perlakuan (A) 15% ; (B) 30% ; (C) 45% ; (D) 60% dan (E) 75%. Masing – masing perlakuan diulang sebanyak 3 kali. Hasil penelitian menunjukan bahwa perlakuan pemberian ekstrak kasar brotowali terhadap bakteri V. harveyi berbeda sangat nyata yaitu pada pengamatan diameter zona bening didapatkan nilai tertinggi pada perlakuan E dengan rata-rata 7,33 mm. Sedangkan diameter rata – rata zona bening terendah pada perlakuan A sebesar 3,63 mm. Hubungan zona bening antar perlakuan ekstrak kasar brotowali terhadap bakteri V. harveyi menunjukan perpotongan garis secara linear dengan persamaan y = 2,7237 + 0,0616x dengan koefisien nilai determinasi (R2) sebesar 0,8482. Hubungan antara pemberian ekstrak kasar brotowali (T. cordifolia) dalam menghambat pertumbuhan bakteri V. harveyi dengan rata-rata diameter zona bening yang dihasilkan menunjukkan respon yang meningkat. Ekstrak kasar brotowali (T. cordifolia) mampu menghambat bakteri V. harveyi. Ekstrak kasar brotowali (T. cordifolia) dapat diaplikasikan sebagai bahan obat namun harus dilakukan uji lebih lanjut agar diketahui dosis optimum dan uji in vivo untuk membuktikan keefektifan bahan tersebut.

English Abstract

The use of antibiotics in cultivation mostly causes of effects, such as raising residues and making bacteria more resistant . This research was aimed to understand the power of resistor the brotowali rough extract (T. cordifolia) against bacteria V. harveyi. The method used in this research is experimental method. The experimental design used was Completely Randomized Design (RAL) using 5 brotowali extract treatment dose (A) 15%, (B) 30%, (C) 45%, (D) 60% and (E) 75%. Each treatment was repeated 3 times. The result of this research showed that the treatment of brotowali extract to V. harveyi bacteria was very significant, in the observation of the clear zone, the highest value was obtained in treatment E with an average of 7,33 mm. While the average of the lowest clear zone in treatment A amounted to 3,63 mm. The correlation of the clear zone between the treatments of brotowali extract to V. harveyi bacteria showed linearly crossed line with the equation y = 2,7237 + 0,0616x with coefficient of determination value (R2) equal to 0,8482. The association between the administration of brotowali crude extract (T. cordifolia) in inhibiting the growth of V. harveyi bacteria with the resulting clear-zone mean showed increased response along with the increase of dose. Brotowali crude extract (T. cordifolia) is able to inhibit V. harveyi bacteria. Brotowali crude extract (T. cordifolia) can be applied as a medicinal substance but should be further tested to determine the optimum dose and in vivo test to prove the effectiveness of the material.
