Abstract
Kerusakan Pada Sel-Sel Beta Penghasil Insulin Menyebabkan Produksi Atau Sekresi Mengalami Penurunan. Keadaan Ini Dapat Menyebabkan Kondisi Hiperglikemia Yang Mengakibatkan Terjadinya Penyakit Diabetes Mellitus. Hiperglikemia Merupakan Suatu Kondisi Dimana Terjadi Peningkatan Kadar Glukosa Melebihi Batas Normal, Dengan Peningkatan Kadar Gula Darah Puasa ≥ 126 Mg/Dl (7.0 Mmol/L) Dan Peningkatan Kadar Gula Sewaktu ≥ 200 Mg/Dl. Tujuan Penelitian Ini Untuk Mengetahui Pengaruh Pemberian Ekstrak Daun Seledri (Apium Graveolens L..) Secara Oral Terhadap Jumlah Sel Beta Pada Mencit Yang Diinduksi Streptozotocin. Metode Penelitian Ini Menggunakan Randomized Posttest Only Controlled Group Design. Hewan Coba Dibagi Dalam Lima Kelompok, Diantaranya: Kelompok Hewan Coba Dalam Kondisi Sehat, Kelompok Dengan Hiperglikemia, Kelompok Hiperglikemia Dengan Pemberian Ekstrak Seledri Sebesar 200 Mg/Kgbb, Kelompok Hiperglikemia Dengan Pemberian Ekstrak Seledri Sebesar 400 Mg/Kgbb, Dan Kelompok Hiperglikemia Dengan Pemberian Ekstrak Seledri Sebesar 800 Mg/Kgbb. Terminasi Dilakukan Pada Hari Ke 60 Setelah Pemberian Terapi Dan Dilakukan Pewarnaan He Pankreas. Penghitungan Jumlah Sel Dilakukan Dengan Foto Slide Perbesaran 400x Dan Diambil 5 Lapang Pandang. Analisa Statistika Dengan Menggunakan Uji One-Way Anova (Α=0,05). Hasil Penelitian Menunjukkan Bahwa Ekstrak Daun Seledri Bepengaruh Terhadap Jumlah Sel Beta Pankreas. Penelitian Selanjutnya Direkomendasikan Menggunakan Pewarnaan Imunohistokimia Dengan Membandingkan Parameter Lain.
English Abstract
Damage Of Beta Cells Producing Insulin Causes Decreased Production Or Secretion. This Situation Can Cause A Condition That Results In Hyperglycemia Of Diabetes Mellitus. Hyperglycemia Is A Condition Where There Is An Increase In Glucose Levels Than Normal, With An Increase In Fasting Blood Glucose Levels ≥ 126 Mg / Dl (7.0 Mmol / L) And Increased Random Blood Glucose Levels As ≥ 200 Mg / Dl.. This Research Method Using A Randomized Controlled Group Posttest Only Design. Experimental Animals Were Divided Into Five Groups, Including: A Group Of Experimental Animals In A Healthy Condition, The Group With Hyperglycemia, Hyperglycemia Group With Celery Extract At 200 Mg / Kg Body Weight, Hyperglycemia Group With Celery Extract At 400 Mg / Kg, And Hyperglycemia Group With Extract Celery Is 800 Mg / Kg. Termination Performed On Day 60 After Therapy And Pancreas He Staining. Counting The Number Of Cells Is Done With A Photo Slide With 400x Magnification In 5 Fields Of View. Statistical Analysis By Using One-Way Anova (Α = 0.05). The Result Suggests The Significant Difference Between Positive Control Group And All Three Treatment Groups. In Conclusion, The Ethanol Extract Of Celery Give Effect To Beta Cell. Thus, It Can Promote As Complementary Therapy For Hyperglycemia. Recommended Further Research Using Immunohistochemical Staining By Comparing Other Parameters.
