Abstract

Penyakit Parkinson menduduki peringkat kedua di dunia setelah Penyakit Alzheimer yang merupakan suatu penyakit saraf degeneratif. Terapi regeneratif untuk Penyakit Parkinson sangat potensial untuk dikembangkan. Salah satu modalitas terapi regeneratif adalah menggunakan Hematopoietic Stem Cells (HSCs) yang berasal dari sum-sum tulang belakang. Hematopoietic Stem Cells memiliki kemampuan untuk meregenerasi jaringan yang rusak dengan cara bergerak menuju jaringan yang rusak. Pergerakan HSCs ini dipicu oleh meningkatnya Granulocyte-colony stimulating factor (G-CSF). Beta glucan pada Saccharomyces cerevisiae dapat meningkatkan jumlah dari G-CSF. Tujuan dari penelitian ini adalah untuk mengetahui adanya perubahan fungsi motorik tikus setalah pemberian Saccharomyces cerevisiae. Penelitian ini menerapkan eksperimen murni (true experimental design) secara in vivo dengan rancangan randomized pre and post test controlled group design. Sampel dibagi ke dalam lima keompok dan masing-masing terdiri dari 5 ekor tikus, yaitu Kelompok Kontrol Negatif, Kelompok Kontrol Positif, Kelompok Perlakuan 1 (Rotenone + Saccharomyces cerevisiae 18 mg/kgBB), Kelompok Perlakuan 2 (Rotenone + Saccharomyces cerevisiae 36 mg/kgBB), Kelompok Perlakuan 3 (Rotenone + Saccharomyces cerevisiae 72 mg/kgBB) selama 30 hari. Variabel yang diukur pada penelitian ini adalah perubahan fungsi motorik dari tikus, yang diukur dengan metode cylinder test. Hasil dari penelitian ini menunjukkan bahwa tidak terdapat perbedaan signifikan fungsi motorik tikus pada kelompok positif terhadap kelompok kontrol negatif baik Pre Test maupun Post Test (p=0,093; 0,093), tetapi terdapat perbedaan signifikan fungsi motorik pada kelompok perlakuan 1, 2 dan 3 terhadap kontrol negatif pada Pre Test (p=0,046; 0,021; 0,026) dan Post Test (p=0,047; 0,020; 0,031). Hasil uji korelasi menunjukkan terdapat korelasi yang cukup kuat (r=0,624) dan arah korelasi positif, sehingga semakin besar dosis Saccharomyces cerevisiae, semakin meningkat perbaikan fungsi motorik tikus. Kesimpulan dari penelitian ini adalah pemberian Saccharomyces cerevisiae mampu memperbaiki fungsi motorik tikus strain Wistar model parkinson secara signifikan, dengan hasil maksimal pada dosis 72 mg/kgBB.

English Abstract

Parkinson’s Disease is a degenerative neurological disease, that is ranked as second most prevalence neurological disease in the world after Alzheimer’s Disease. The regenerative therapy for Parkinson’s Disease is so potential to be developed. One of the regenerative therapeutic modality is using Hematopoietic Stem Cells (HSCs) derived from the bone marrow. Hematopoeitic Stem Cells has the ability to regenerate the damaged tissue by moving towards the damaged parts and regenerate the cell. This HSCs movement is triggered by the increasing of Granulocyte-Colony Stimulating Factors (G-CSF). Beta glucan in Saccharomyces cerevisiae can increase the amount of G-CSF. The purpose of this research was to investigate wether there is any change of motoric function after the administration of Saccharomyces cerevisiae in Parkinson’s rat model. This research was applied a true experimental design by in vivo with draft randomized controlled pre test and post test group design. The samples is divided into five groups and each of them consists of 5 rats, namely, Negative Control Group, Positive Control Group, Treatment Group 1 (Rotenone + Saccharomyces cerevisiae 18 mg/kg), Treatment Group 2 (Rotenone + Saccharomyces cerevisiae 36 mg/kg), Treatment Group 3 (Rotenone + Saccharomyces cerevisiae 72 mg/kg) for 30 days. The variables that is measured in this research is the change of the motoric function of rats, it is measured by the cylinder test method. The results of this research shows that there is no significant difference in the Positive Control Group of rats motoric function to the Negative Control Group on both Pre and Post Test (p=0,093; 0,093), but there is significant differences in motoric function in the Treatment Group 1, 2 and 3 to Negative Control Group on the Pre Test (p=0,046; 0,021; 0,026) and Post Test (p=0,047; 0,020; 0,031). The results of correlation test results shows a fairly strong correlation (r=0,624) and a positive correlation direction too. It means, the highest dose of Saccharomyces cerevisiae, more increasing improvement of motoric function of rats. The conclusion of this research is the administration of Saccharomyces cerevisiae is able to improve the motoric function of Parkinson’s Wistar strain rat model significantly, with maximum results at 72 mg/kg dose.
