Abstract
Amoebiasis adalah suatu keadaan terdapatnya Entamoeba histolytica dengan atau tanpa manifestasi klinik dan disebut sebagai Food Borne Disease. Staphylococcus aureus memiliki kemiripan struktur protein yang sama dengan Entamoeba histolytica yaitu Gal/GalNAc lectin yang secara spesifiknya merupakan rekombinan protein pada Gal/GalNAc lectin yang disebut Lec A. Tujuan penelitian ini untuk memperoleh bukti bahwa Lec A bakteri Staphylococcus aureus dapat meningkatkan kadar IgG pada keadaan amoebiasis yang disebabkan oleh Entamoeba Histolytica. Penelitian ini menggunakan desain eksperimen murni secara in vivo menggunakan rancangan Randomized Pre-Post Test Controlled Group Design. Tikus wistar sebagai hewan coba dibagi menjadi 5 kelompok, yaitu: Kontrol Negatif (tanpa perlakuan), Kontrol Positif (induksi trophozoite Entamoeba histolytica), P1 (Lec A 0,1cc/KgBB + induksi trophozoite Entamoeba histolytica); P2 (Lec A 0,15cc /KgBB + induksi trophozoite Entamoeba histolytica); P3 (Lec A 0,2cc/KgBB + induksi trophozoite Entamoeba histolytica). Pengukuran IgG menggunakan metode ELISA. Dari hasil penelitian diperoleh hubungan yang signifikan antara kadar IgG dengan dosis Lec A setelah masa vaksinasi dan setelah induksi Entamoeba histolytica. Dosis Lec A 0.2 cc/KgBB secara signifikan mampu meningkatkan IgG. Kesimpulan dari penelitian ini adalah Lec A bakteri Staphylococcus aureus terbukti mampu meningkatkan IgG tikus wistar.
English Abstract
Amoebiasis is a state of the presence of Entamoeba histolytica with or without clinical manifestations and Food Borne Disease called. Staphylococcus aureus has the same protein structure similarity with Entamoeba histolytica is Gal/GalNAc lectin which is specifically a recombinant protein in the Gal/GalNAc lectin called Lec A. The purpose of this study to obtain evidence that Lec A Staphylococcus aureus bacteria can increase the levels of IgG in amoebiasis circumstances caused by Entamoeba histolytica. This study uses a pure experimental design in vivo using randomized designs Pre-Post Test Controlled Group Design. Wistar rats as experimental animals were divided into 5 groups: negative control (without treatment), Positive Control (Entamoeba histolytica trophozoite induction), P1 (Lec A 0,1cc/KgBW + Entamoeba histolytica trophozoite induction); P2 (Lec A 0,15cc/KgBW + Entamoeba histolytica trophozoite induction); P3 (Lec A 0.2cc/KgBW + induction Entamoeba histolytica trophozoite). Measurement of IgG using the ELISA method. The result showed a significant relationship between dose levels of IgG with Lec A after vaccination period and after induction of Entamoeba histolytica. Lec A dose of 0.2cc/KgBW can significantly improve IgG. The conclusion of this study is Lec A Staphylococcus aureus proved to increase the wistar rat IgG.
