Abstract

Malaria Merupakan Penyakit Infeksi Menular Yang Sampai Sekarang Masih Menjadi Masalah Kesehatan Dunia. Indonesia Merupakan Salah Satu Negara Yang Beresiko Tinggi Terhadap Penyakit Malaria Dan Penyakit Ini Masih Ditemukan Di Seluruh Provinsi Di Indonesia. Hal Ini Di Perparah Dengan Terjadinya Resistensi Obat Antimalaria Lini Pertama Yaitu Artemisinin Combination Therapy (Act) Serta Belum Adanya Vaksin Anti-Malaria Efektif Yang Tersedia Untuk Digunakan Pada Manusia Sehingga Sangat Diperlukan Penemuan Obat/Terapi Baru Untuk Malaria. Ubiquitin Proteasom System (Ups) Berperan Dalam Mengontrol Kualitas Protein, Degradasi Protein Serta Proliferasi Sel. Pada Plasmodium Sp. Ups Yang Diperankan Oleh Proteasome 20s Memiliki Peran Penting Dalam Mengontrol Kualitas Protein. Ubiquitin Proteasom System (Ups) Merupakan Target Baru Yang Potensial Untuk Terapi Malaria Karena Penghambatan Pada Core Protein Proteasome 20s Dapat Menghambat Pertumbuhan Plasmodium Sp. Penelitian Ini Merupakan Penelitian Murni Dengan Metode Randomized Posttest Only Controlled Group Design. Antigen Proteasome 20s Didapatkan Melalui Proses Ekstraksi Dari Plasmodium Berghei Yang Diinokulasikan Pada Mencit Hidup. Mencit Yang Dijadikan Hewan Coba Dibagi Menjadi 6 Kelompok Yaitu: Kontrol Positif, Kontrol Negatif, Ag-Proteasom 1µl + Cfa-Ifa 100µl, Ag-Proteasom 2µl + Cfa-Ifa 100µl, Ag-Proteasom 4µl + Cfa-Ifa 100µl, Dan Ag-Proteasom 8µl + Cfa-Ifa 100µl. Seluruh Kelompok Mencit Kecuali Kontrol Negatif Diinfeksi Dengan Plasmodium Berghei Setelah Diberikan Perlakuan. Hasil Penelitian Menunjukkan Terdapat Perubahan Morfologi Parasit Menjadi Crisis Form Pada Kelompok Kontrol Positif. Pemberian Antigen Proteasome 20s Mampu Memperbaiki Morfologi Parasit Dalam Pengamatan Tiap Hari Selama 4 Hari.

English Abstract

Malaria Is An Infectious Disease That Still Being A World Health Problem. Indonesia Is The One Of Tropical Countries That Has A High Risk For Malaria Infection And The Disease Can Be Found In All Provinces Of Indonesia. It Is In Aggravated By The Drug Resistance In Artemisinin Combination Therapy (Act), A First-Line Antimalarial And The Lack Of An Effective Anti-Malaria Vaccine For Use In Humans. Therefore It Is Indispensable For Drug Discovery Or New Therapies For Malaria. Ubiquitin Proteasome System (Ups) Plays A Role In Controlling The Quality Of The Protein, Protein Degradation And Cell Proliferation. In Plasmodium Sp. Ups That Played By 20s Proteasome Has An Important Role In Controlling The Quality Of The Protein. Ubiquitin Proteasome System (Ups) Is A Potential New Target For Malaria Therapy As Inhibition Of 20s Proteasome Core Protein Can Inhibit The Growth Of Plasmodium Sp. This Study Is Purely With The Method Posttest Only Randomized Controlled Group Design. 20s Proteasome Antigen Was Obtained Through Extraction Of Plasmodium Berghei From Infected Mice. Mice Were Used As Experimental Animals Were Divided Into 6 Groups: Positive Control, Negative Control, Ag-Proteasome 1μl + Cfa-Ifa 100μl, Ag-Proteasome 2μl + Cfa-Ifa 100μl, Ag-Proteasome 4μl + Cfa-Ifa 100μl, And Ag- Proteasome 8μl + Cfa-Ifa 100μl. The Entire Group Of Mice Except The Negative Control Infected With Plasmodium Berghei After Given A Treatment. The Results Showed There Is A Change In The Morphology Of The Parasite Into Crisis Form In The Positive Control Group. Administration Of 20s Proteasome Antigen Was Capable In Preventing Morphological Damage Of Parasites In 4 Days Observation
