Abstract
Kanker kolon adalah keganasan dari sel epitel usus besar. Kolitis ulseratif meningkatkan resiko kejadian kanker kolon. Salah satu sitokin pro inflamasi yang berperan adalah tumor necrosis factor-α (TNF-α). Peranan limpa dalam patogenesis kolitis yaitu meningkatkan jumlah sel imun. Tanaman yang dapat digunakan sebagai pendamping pengobatan kanker kolon adalah benalu mangga (Dendropthoe pentandra) karena mengandung kuersetin. Tujuan penelitian ini untuk menunjukkan pemberian ekstrak etanol daun benalu mangga (Dendropthoe pentandra) dapat menurunkan ekspresi gen TNF-α pada limpa mencit model colitis-associated colon cancer. Penelitian ini menggunakan true experimental post-test only control group design yang terdiri dari 5 kelompok. Semua kelompok diberi AOM intraperitoneal 10mg/kgBB pada hari pertama, kemudian DSS 5% per oral dicampur air minum pada minggu ke 1,3,5,7,9,11, dan 13, kecuali kelompok E (kontrol negatif). Ekstraketanol daun benalu mangga (Dendropthoe pentandra) diberikan dengan konsentrasi 125mg/kgBB, 250 mg/kgBB, D 500 mg/kgBB. Pada minggu ke 15 mencit dikorbankan untuk dilihat gen TNF-α pada limpa. Hasil uji statistik menunjukkan perbedaan gen TNF-α yang signifikan antara kelompok kontrol dan perlakuan (p<0,05), uji korelasi antar variabel menunjukkan korelasi yang kuat (p=0,000) dan koefisien korelasi negatif (-0,963). Kesimpulan penelitian ini ekstrak etanol daun benalu mangga (Dendropthoe pentandra) dapat menurunkan gen TNF-α pada limpa mencit model colitis-associated colon cancer dengan dosis optimal 500 mg/kgBB.
English Abstract
Colon cancer is malignancy of the colon epithelial cells. Ulcerative colitis increase the risk of incidence in colon cancer. One of the pro-inflammatory cytokine that play a role is tumor necrosis factor-α (TNF-α). Spleen role in colitis pathogenesis is the upregulation of immune cell. Plants that can be used as adjuvant to colon cancer treatment is Dendropthoe pentandra because it contain quercetin. This study aims to demonstrate that the Dendropthoe pentandra leaf ethanol extract can reduce the gene of TNF-α in spleen mice model colitis-associated colon cancer. This study used true experimental post test only control group design which consist of 5 groups. All of them were given 10 mg of AOM intraperitoneal on first day, then 5% of DSS orally mixed in drinking water in 1st, 3rd,5th,7th,9th,11th, and 13th, except for group E (control negative). Dendropthoe pentandra leaf ethanol extract is given with concentration 125 mg/kgBW, 250 mg/kgBW, 500 mg/kgBW. At 15th week, mice were sacrificed to look for TNF-α gene in spleen. Statistic studies result showed TNF-α gen differences were significant between control and treatment group (p<0,05), correlation study between variables showed a strong correlation (p=0,000) and negative correlation coefisien (-0,963). This study conclusion is that Dendropthoe pentandra leaf ethanol extract can reduce TNF-α gen in spleen mice model colitis-associated colon cancer with optimal doses 500 mg/kg BW.
