Abstract
Kanker kolon dapat terjadi karena proses inflamasi kronis di kolon yang biasa disebut Colitis Associated Colon Cancer (CAC). Kondisi tersebut dipengaruhi oleh aktivasi jalur cyclooxygenase-2 (COX-2), inflamasi kronis serta agen-agen proinflamasi yang semakin memperparah inflamasi, merusak jaringan mukosa kolon, hingga mengarah pada keadaan prakanker atau displasia. Daun benalu mangga (Dendrophthoe pentandra) mengandung flavonoid yang tinggi terutama kuersetin sebagai antiinflamasi penghambat COX-2. Mekanisme kerja tersebut yang diharapkan mampu menghambat keberlanjutan proses inflamasi dinilai dengan indeks kerusakan histologi dan proporsi displasia. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol daun benalu mangga terhadap penurunan Indeks kerusakan histologi dan displasia pada mencit model CAC. Penelitian eksperimental laboratoris ini menggunakan metode randomized post test only controlled group design. Penelitian ini menggunakan 20 ekor mencit yang dikelompokkan menjadi 5 kelompok: 2 kelompok Kontrol (positif dan negatif) dan 3 kelompok dengan dosis 0,125 mg/gBB; 0,25 mg/gBB; dan 0,5 mg/gBB. Setelah AOM dan DSS diinduksikan, dosis diberikan mulai minggu ke-6 sampai minggu ke-14. Pada minggu ke-15 dilakukan pembedahan, pembuatan preparat histologi kolon dengan pewarnaan HE dan dianalisa di bawah mikroskop untuk menilai indeks kerusakan histologi dan displasia yang terjadi. Hasil ANOVA menunjukkan hasil indeks kerusakan histologi yang signifikan, p = 0.004 (p < 0.05). Hasil proporsi displasia juga menunjukkan adanya penurunan proporsi dari kontrol positif ke kelompok berikutnya. Dapat disimpulkan bahwa pemberian ekstrak etanol daun benalu mangga mampu menurunkan indeks kerusakan histologi dan displasia pada mencit model CAC.
English Abstract
Colon cancer can occurs caused by a chronic inflammatory process in colon and commonly called Colitis Associated Colon Cancer (CAC). These conditions are influenced by the activation of cyclooxygenase-2 (COX-2) pathway, chronic inflammation and proinflammatory agents which further aggravate the inflammation, destroy mucosa tissue and leading to pracancer condition or dysplasia. Dendrophthoe pentandra’s leaves contains abundant flavonoids especially quercetin as an antiinflammatory inhibit COX-2 pathway. That mechanism which is expected impair the continous of inflammation process assessed using histological damage index and dysplasia proportion. This study aimed to determine the effect of ethanol extract of Dendrophthoe pentandra’s leaves for histological damage index and dysplasia in CAC mice model.This laboratory experimental study using randomized post test only controlled group design. This study used 20 mice that divided into 5 groups: 2 control groups (positive and negative) and 3 groups received 0,125 mg/gBW; 0,25 mg/gBW; and 0,5 mg/gBW. After induced AOM and DSS, the doses were given from 6th week to 14th week, at 15th week then mice were sacrificed, made colon histological slide with HE staining and analyzed under a microscope to assess the histological damage index and dysplasia. ANOVA’s result showed the histological damage index significantly, p = 0.004 (p <0.05). Results of dysplasia proportion also showed decrease proportion from positive control group to the next group. It could be concluded that the ethanol extract of Dendrophthoe pentandra’s leaves were able to reduce the histological damage index and dysplasia in CAC mice model.
