Abstract

Infeksi malaria pada kehamilan dapat menyebabkan kematian maternal dan berat badan lahir rendah pada fetus. Hal ini berhubungan dengan pengaruh dari peningkatan mediator inflamasi dan peningkatan aktivitas sel imun. Penelitian ini bertujuan untuk mengetahui peran dari limfosit T CD4 pada proses patologi perkembangan fetus pada kehamilan mencit Balb/C dengan infeksi Plasmodium Berghei . Mencit balb/c betina (primigravida) dibagi menjadi kelompok perlakuan (9 mencit diinfeksi Plasmodium berghei pada hari ke-9 kehamilan) dan kelompok kontrol (8 mencit normal). Pembedahan dilakukan pada hari ke-18 kehamilan untuk mengisolasi fetus dan plasenta. Peningkatan ekspresi limfosit T CD4 pada plasenta dihitung dengan pewarnaan immunohistokimia dan berat badan janin diukur dengan skala analitikal (Mettler AE 50). Pada uji Independent T test menunjukkan ekspresi limfosit T CD4 (p=0.009), dan berat badan janin (p=0.003) meningkat pada kelompok perlakuan. Pada uji korelasi menunjukkan adanya korelasi negatif antara ekspresi limfosit T CD4 (r=-0.665, p=0.004) dengan berat badan janin. Kesimpulan dari penelitian ini adalah peningkatan ekspresi limfosit T CD4 memiliki peran penting dalam patogenesis dari berat badan janin yang rendah pada kehamilan dengan infeksi malaria

English Abstract

Malarial infection during pregnancy can cause maternal mortality and low birth weight of the fetal. Those can be influenced by elevating of mediator inflammatory and increasing of immune cells activities. This study was aimed to know the role of CD4 lymphocyte T cells in pathological process of fetal development in pregnant Plasmodium berghei infected BALB/c mice. Female Mus musculus BALB/c (primigravida) were divided into positive control group (9 mice infected by Plasmodium berghei on the 9 th day of pregnancy) and negative control group (8 normal mice). Surgery performed on the 18 th day of pregnancy to isolate the fetus and the placenta. Expression of CD4 lymphocyte T cells on placenta was counted by immunohistochemical staining and fetal weights were measured by analytical scale (Mettler AE 50). Independent T test showed that expression of CD4 lymphocyte T cells (p=0.009), and fetal weight (p=0.003) increase significantly in positive control group. Moreover, Pearson correlation test showed that there was a negative correlation between expression of CD4 lymphocyte T cells (r=-0.665, p=0.004) and fetal weight. The conclusion of this study is that elevating of CD4 lymphocyte T cells have an important role in the pathogenesis of low fetal weight during malaria infection.
