Uji Daya Antihelmintik Ekstrak Etanol Bunga Kembang Sepatu (Hibiscus rosa-sinensis L) Terhadap Cacing Ascaris suum Secara In Vitro

Abstract

Askariasis adalah penyakit cacingan yang paling besar prevalensinya di antara penyakit cacing lainnya di Indonesia. Obat cacing yang menjadi pilihan terhadap askariasis adalah piperazin, pirantel pamoat, albendazole atau mebendazol. Namun, ketiga obat tersebut memiliki efek samping berupa gangguan saluran pencernaan berupa sakit perut dan diare serta kontraindikasi pada wanita hamil karena memiliki efek teratogen. Usaha alternatif sebagai antihelmintik askariasis yaitu dengan menggunakan ekstrak bunga kembang sepatu. Bunga kembang sepatu mengandung flavonoid dan saponin yang merupakan zat yang memiliki sifat antihelmintik. Tujuan penelitian ini adalah untuk mengetahui seberapa besar daya antihelmintik dari ekstrak bunga kembang sepatu (Hibiscus rosa-sinensis L) terhadap cacing Ascaris suum secara in vitro. Penelitian ini merupakan penelitian eksperimental laboratoris dengan rancangan true experimental-post test only control group design. Untuk menentukan cacing Ascaris suum yang diikutkan dalam penelitian ini maka digunakan teknik kriteria inklusi dan eksklusi. Penelitian ini menggunakan ekstrak bunga kembang sepatu (Hibiscus rosa-sinensis L) dengan konsentrasi 20%, 30% dan 40% serta FBS 1% dalam larutan PBS sebagai kontrol negatif dan pirantel pamoat 1% sebagai kontrol positif yang di ulang sebanyak empat kali yang di lihat pada interval waktu yaitu pada jam 1,2,3,4,5,6,7,8 dan 24. Pirantel pamoat digunakan sebagai kontrol positif karena merupakan terapi lini pertama dari askariasis itu sendiri. Hasil penelitian menunjukkan bahwa konsentrasi 40% pada waktu 24 jam dapat membunuh 100% cacing. Nilai signifikansi antara konsentrasi ektrak bunga kembang sepatu terhadap daya antihelmintik menunjukkan bahwa hubungan antara konsentrasi ekstrak bunga kembang sepatu terhadap daya antihelmintik adalah bermakna (p<0,05), dimana di dapatkan p-value 0.410 artinya semakin tinggi konsentrasi ekstrak maka semakin besar jumlah cacing yang mati. Dapat disimpulkan bahwa ekstrak bunga kembang sepatu (Hibiscus rosa-sinensis L) memiliki daya antihelmintik terhadap cacing Ascaris suum.

English Abstract

Ascariasis is one of the high prevalence worm infections in Indonesia. The usual antihelmintics prescribed are piperazine, pyrantel pamoate, albendazole or mebendazole. However, these three drugs have side effects such as gastrointestinal disorders. Examples of such side effects are abdominal pain and diarrhea. These drugs are contraindicated in pregnant women because it has teratogenic effect. Alternate anthelmintics for ascariasis is by using hibiscus flower extract. Hibiscus flower contains flavonoids and saponins which have properties of anthelmintics. The purpose of this study is to determine the potency of anthelmintics from hibiscus extract (Hibiscus rosa-sinensis L) against Ascaris suum. The study was a laboratory experimental research with true experimental design-post test only controls group design. Inclusion and exclusion technique criteria were used to determine which Ascaris suum that will be used in this study. The research used hibiscus flower extract (Hibiscus rosa-sinensis L) with concentrations of 20%, 30% and 40%. 1% of FBS in PBS solution act as a negative control while 1% pyrantel pamoate act as a positive control. The experiment were repeated four times and each repetition was observed at an interval (1st hour, 2nd hour, 3rd hour, 4th hour, 5th hour, 6th hour, 7th hour, 8th hour, and 24th hour). Pyrantel pamoate was used as a positive control because it is the first-line therapy of ascariasis. The results showed that the concentration of 40% at 24 hours killed 100% of ascaris. Significance value between hibiscus flower extract concentration against anthelmintics which means that the relationship between concentration of hibiscus flower extract as an anthelmintics are significant (p<0.05). The p-value 0.410 was obtained, which means higher the concentration of the extract, the greater the number of dead ascaris. Based on this result, we can conclude that the extract of hibiscus flower (Hibiscus rosa-sinensis L), has anthelmintic properties against Ascaris suum.
