Abstract

Demam tifoid termasuk penyakit endemik yang menular melalui makanan dan minuman yang terkontaminasi oleh bakteri Salmonella Typhi yang saat ini resisten terhadap sebagian besar antibiotik lini pertama. Eksplorasi dilakukan untuk mencari antibakteri yang efektif, salah satunya dengan memanfaatkan buah sawo mentah ( Manilkara zapota ) yang memiliki kandungan tannin, flavonoid dan saponin yang dipercaya mempunyai efek antibakteri. Tujuan penelitian ini yaitu untuk mengetahui efek antibakteri ekstrak buah sawo mentah ( Manilkara zapota ) terhadap pertumbuhan bakteri Salmonella Typhi secara in vitro . Sampel bakteri yang digunakan adalah bakteri Salmonella Typhi yang dimiliki oleh Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Brawijaya Malang yang berasal dari darah pasien Rumah Sakit Saiful Anwar (RSSA). Konsentrasi ekstrak yang dipakai adalah 6%, 5%, 4%, 3%, 2%, 1% dan 0%. Metode penelitian yang digunakan yaitu dilusi agar. Metode analisis yang digunakan yaitu Kruskal Wallis dengan p= 0,000. Hasil analisis Mann Whitney menunjukkan terdapat perbedaan yang signifikan antara perubahan konsentrasi ekstrak buah sawo mentah terhadap pertumbuhan bakteri Salmonella Typhi. Uji Korelasi Spearman menunjukkan dengan peningkatan konsentrasi ekstrak buah sawo mentah menyebabkan penurunan pertumbuhan bakteri Salmonella Typhi, r= -0,946. KHM ekstrak bua sawo mentah terhadap bakteri Salmonella Typhi didapatkan pada konsentrasi 5%. Kesimpulan pada penelitian ini yaitu buah sawo mentah mempunyai efek antibakteri terhadap bakteri Salmonella Typhi secara in vitro .

English Abstract

Typhoid fever is an endemic disease that is transmitted through food, drinks and water contaminated by Salmonella Typhi which resistant to most of first line antibiotics. Many researches have been done to find an effective antibacterial, one of these is by utilizing unripe Sapota ( Manilkara zapota ) fruit contains tannin, flavonoid and saponin, is believed to have an antibacterial activity. This research aimed to determine the antibacterial effect of unripe Sapota ( Manilkara zapota ) fruit extract on Salmonella Typhi in vitro. Sample of bacteria was obtained from clinical isolates provided by the Microbiology Laboratory of the Medical faculty of Brawijaya University which is taken from patient blood samples of Saiful Anwar Hospital. The extract concentrations were 6%, 5%, 4%, 3%, 2%, 1% and 0%. The used method was agar dilution method. The data analysis was based on the Kruskal Wallis method with p= 0,000. The data analysis from Mann Whitney method showed significant differences with the different concentrations of unripe Sapota ( Manilkara zapota ) fruit extract to the Salmonella Typhi growth. The correlation Spearman test showed that the higher extract concentration causes the lower Salmonella Typhi growth, r= -0,946. Minimum Inhibitory Concentration (MIC) of unripe Sapota ( Manilkara zapota ) fruit extract against Salmonella Typhi is observed at the concentration 5%. The conclusion of this research was that unripe Sapota ( Manilkara zapota ) fruit extract has antibacterial effect on Salmonella Typhi in vitro .
