Abstract
Fibrosis hati merupakan masalah kesehatan yang banyak menimbulkan dampak kematian. Sebagian besar kematian disebabkan oleh stadium akhir dari fibrosis itu. Hal ini disebabkan karena fibrosis hati merupakan keadaan yang jarang disadari karena tidak menimbulkan gejala dalam kurun waktu lama. Padahal, fibrosis hati merupakan keadaan yang reversible dan masih bisa diobati. Oleh karena itu, screening dan penentuan derajat fibrosis sangat diperlukan untuk mencegah perkembangan fibrosis hati. Di lain sisi, IL-17, yang disekresikan oleh sel Th-17, pada keadaan inflamasi kronis diprediksi berkontribusi terhadap proses fibrosis hati. Penelitian ini bertujuan untuk mengetahui hubungan antara ekspresi IL-17 pada limfosit jaringan hati dengan derajat fibrosis hati pada tikus yang diinduksi karbon tetraklorida (CCl4) dosis 1 mL/kgBB dengan lama pemaparan 2 minggu, 5 minggu, dan 9 minggu, untuk mendapat derajat fibrosis yang berbeda. Penelitian ini merupakan penelitian eksperimental dengan metode Randomized Post Test Only Control Group. Hewan coba dibagi menjadi empat kelompok perlakuan, dengan derajat fibrosis yang berbeda yaitu fibrosis derajat 0, derajat 1, derajat 2, dan derajat 3. Diakhir perlakuan, ekspresi IL-17 pada limfosit jaringan dan derajat fibrosis hati dibandingkan. Dari hasil penelitian didapatkan rata-rata ekspresi IL-17 pada limfosit jaringan hati senilai 0,34 pada kelompok kontrol, 0,69 pada kelompok fibrosis derajat 1, 2,77 pada kelompok fibrosis derajat 2, dan 4,98 pada kelompok fibrosis derajat 3. Sehingga didapat kesimpulan, terdapat hubungan korelasi positif antara ekspresi IL-17 pada limfosit jaringan hati dengan derajat fibrosis hati dengan p=0,083.
English Abstract
Liver fibrosis is one of the health problem which has a big number of events leading to death. Most of the death incidents are caused by the end stage of the liver fibrosis. This phenomenon is because of the fact that liver fibrosis is usually asymptomatic. However, liver fibrosis is a reversible and curable condition. Because of that, screening and determination of the fibrosis stage is needed to prevent the development of fibrosis. On the other side, IL-17, secreted by Th-17 cell, in a recurrent exposure of injury predicted to contribute to the fibrosis process. This research aim to know the correlation between IL-17 liver tissue lymphocytes expression with the stages of liver fibrosis in rats induced by carbon tetrachloride(CCl4) 1 mL/kgBB with different exposure terms (2 weeks, 5 weeks, and 9 weeks), to get diferrent stages of liver fibrosis . This is an experimental research with Randomized Post Test Only Control Group method. The experimental rats divided into 4 groups differ by the fibrosis stage, which are, fibrosis stage 0, stage 1, stage 2, and stage 3. At the the end of the research, the expression of IL-17 in liver tissue lymphocytes is compared to the stages of liver fibrosis. The result from this research is that there is an increase in IL-17 expression as, 0,34 in control group, 0,69 fibrosis stage 1 group, 2,77 in fibrosis stage 2 group, and 4,98 in fibrosis stage 3 group. It is concluded, that there is a positive correlation between the expression of IL-17 in liver tissue and the stages of liver fibrosis with p = 0,083.
