Abstract

Staphylococcus aureus merupakan salah satu penyebab infeksi nosokomial di rumah sakit pada luka operasi dan infeksi yang terkait dengan indwelling medical devices. Bakteri ini sulit diatasi karena sering menimbulkan rekurensi dan resistensi antibiotika. Faktor penting yang amat berperan adalah kemampuannya membentuk biofilm. Biofilm merupakan suatu agregat mikroba yang melekat pada permukaan substrat. Tujuan penelitian ini adalah untuk mengetahui efek pemberian ekstrak etanol biji pare yang mengandung alkaloid, tannin, flavonoid, dan terpenoid dalam menghambat pembentukan biofilm Staphylococcus aureus. Penelitian menggunakan post-test only control group design pada bakteri Staphylococcus aureus pembentuk biofilm yang dibagi dalam 8 kelompok, yaitu kontrol kuman (tidak diberi ekstrak biji pare), bakteri dengan biji pare dosis 0,00244141%, 0,0046875%, 0,009375%, 0,01875%, 0,0375%, 0,075% dan 0,15% dan dilakukan pengulangan sampai 8 kali. Biofilm diukur Optical Density-nya dengan menggunakan micro plate reader. Analisis data menggunakan metode One way Anova dilanjutkan uji Post Hoc LSD, dan Uji Korelasi Pearson. Hasil penelitian menunjukkan bahwa pemberian ekstrak biji pare memberikan pengaruh dalam menghambat pembentukan biofilm secara signifikan (p<0,05) mulai dari dosis 0.00244141% hingga 0.075% dengan kekeatab korelasi r=-0,116. Kesimpulan penelitian ini adalah pemberian ekstrak etanol biji pare (Momordica charantia) dapat menghambat pembentukan biofilm pada bakteri Staphylococcus aureus.

English Abstract

Staphylococcus aureus is one of cause of nosocomial infections in hospitals and surgical site infections associated with indwelling medical devices. These bacteria are usually difficult to overcome because they often cause recurrence and antibiotic resistance. Important contributing factor is their ability to form biofilms. Biofilm is a microbial aggregate which is attached to the substrate surface. The study aimed to investigate the effect of ethanol extract of bitter melon seed containing alkaloid, tannins, flavonoids, and terpenoids to inhibit biofilm formation of Staphylococcus aureus. This study was using post-test only control group design on biofilm-forming bacteria Staphylococcus aureus which were divided into 8 groups. The control group was bacteria not given the bitter melon seed extract, while the treatment group was given a dose of bitter melon seed extract at doses 0.00244141%, 0.0046875%, 0.009375%, 0.01875%, 0.0375%, 0.075% dan 0.15%. The experiment was repeated up to 8 times. The optical density of biofilm was quantitatively measured using the micro plate reader. Data were analyzed using One way Anova followed by Post Hoc LSD, and the Pearson correlation test. The result showed that bitter melon seed extract significantly (p<0.05) inhibited biofilm formation at doses ranging from 0.00244141% to 0.15% with correlation value r=-0,116, and there was correlation between the dose and the formation of biofilm at Staphylococcus aureus (p=0.000). So, the ethanol extract of bitter melon (Momordica charantia) seed can inhibit biofilm formation in Staphylococcus aureus.
