Abstract
Klebsiella pneumoniae merupakan salah satu bakteri yang mampu memproduksi enzim ESBL ( Extended Spectrum Beta Lactamase ) yang dapat melumpuhkan kerja berbagai jenis antibiotik . Penyebaran infeksi bakteri Klebsiella pneumoniae dapat terjadi di rumah sakit maupun di komunitas, oleh karena bakteri ini sangat mudah menular melalui udara. Sedikitnya pilihan terapi untuk mengobati infeksi Kebsiella pneumoniae membuka peluang penelitian terapi Klebsiella pneumoniae menggunakan bahan alami. Daun sambiloto mengandung berbagai komponen aktif seperti flavonoid, saponin dan tannin yang diduga dapat berperan sebagai antibakteri terhadap Klebisiella pneumoniae . Penelitian ini bertujuan untuk mengetahui efek antimikroba ekstrak etanol daun sambiloto terhadap Klebsiella pneumoniae secara in vitro . Desain penelitian ini adalah desain eksperimental laboratorium. Ekstrak etanol menggunakan metode maserasi dengan pelarut etanol 96%. Metode dilusi tabung digunakan untuk mengetahui Kadar Hambat Minimal (KHM) dan Kadar Bunuh Minimal (KBM). Konsentrasi ekstrak yang digunakan yaitu 18,5%, 17,5%, 16,5%, 15,5%, dan 14,5%.Pada penelitian ini KHM tidak dapat ditentukan karena semua tabung tetap keruh dan KBM berada pada konsentrasi ekstrak 18,5%. Hasil penelitian menunjukkan secara signifikan bahwa peningkatan konsentrasi ekstrak daun sambiloto menyebabkan penurunan tingkat pertumbuhan Klebsiella pneumoniae . Kesimpulan dari penelitian ini adalah ekstrak etanol daun sambiloto memiliki efek antimikroba terhadap Klebsiella pneumoniae secara in vitro .
English Abstract
Klebsiella pneumoniae is one of bacteriae that can produce the ESBL (Extended Spectrum Beta Lactamase) enzim which can deactivate the activity of various types of antibiotics. The transmission of Klebsiella pneumoniae bacterial infection can occur in hospital and in the community, because these bacteriae are very easily transmitted through the air. Few of therapeutic options to treat Klebsiella pneumoniae infection gives opportunities to study Klebsiella pneumoniae treatment using natural ingredients. Sambiloto leaf contains active compounds like flavonoid, saponin and tannin which expected can act as antibacterial to Klebsiella pneumoniae . This study is aimed to know antimicrobial effect of ethanol extract of sambiloto leaf towards Klebsiella pneumoniae in vitro . This study is using laboratory experimental design. Ethanol extract is acquired from maseration method with 96% ethanol as solvent. Tube dilution method is used to observe Minimum Inhibition Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Extract concentration used is 18,5%, 17.5%, 16,5%, 15,5%, and 14,5%. In this study MIC can not be determined because of turbidity in all tubes and MBC on 18,5%. This study result shows significantly that increase of extract concentr ation of sambiloto leaf cause decrease of Klebsiella pneumoniae growth. Conclusion of this study is that ethanol extract of sambiloto leaf has antimicrobial effect towards Klebsiella pneumoniae in vitro .
