Abstract

Hsp27 terekspresi secara berlebihan pada kanker payudara sebagai respon terhadap kondisi stres. Ekspresi Hsp27 yang berlebihan dapat menghambat apoptosis dan mencegah aktivasi caspase. Ekstrak daun kelor (Moringa oleifera) memiliki kandungan flavonoid yang memiliki aktivitas antikanker. Penelitian ini dilakukan untuk mengetahui dosis IC50, pengaruh ekspresi Hsp27 dan apoptosis pada sel MCF-7 oleh ekstrak daun kelor. Studi eksperimental dengan post-test only design dilakukan pada sel MCF-7. Serbuk daun kelor dimaserasi menggunakan etanol 70%. MTT Assay dilakukan pada MCF-7 yang diberi ekstrak daun kelor dengan dosis 7.813-20000 μg/ml. Sel diberikan ekstrak daun kelor pada dosis 1100, 2200 dan 4400 μg/ml dan kemudian dilakukan uji ekspresi Hsp27 (uji imunositokimia) dan apoptosis (TUNEL assay). Selanjutnya, ditentukan indeks ekspresi Hsp27 dan apoptosis, kemudian dilakukan analisis data. Hasil penelitian menunjukkan bahwa IC50 ekstrak daun kelor adalah 2200 μg/ml. Ekstrak daun kelor mampu menurunkan ekspresi Hsp27 secara signifikan pada dosis 2200 dan 4400 μg/ml (p<0.05). Ekstrak daun kelor mampu meningkatkan apoptosis sel MCF-7 secara signifikan pada dosis 1100, 2200, dan 4400 μg/ml (p<0.05). Ekstrak daun kelor sebagai antikanker dapat menurunkan ekspresi Hsp27 dan meningkatkan apoptosis sel kanker payudara MCF-7.

English Abstract

Hsp27 is overexpressed in breast canceras a response to stress condition. Hsp27 overexpression leads to apoptosis inhibition and caspase activation prevention. Drumstick leaves extract (Moringa oleifera) contains flavonoids which have anticancer activity. This research was conducted to find out IC50, effect on Hsp27 expression and apoptosis in MCF-7 cells by drumstick leaves extract. Experimental studies with post-test only design was done on MCF-7 cells. Drumstick leaves powder was macerated with ethanol 70%. MTT Assaywas performed on mcf cell which were treated bydrumstick leaves extract (7.813-20000 μg/ml). Furthermore,drumstick leaves extract treated cells (1100, 2200, and 4400 μg/ml)were assayed for its Hsp27 expression (immunocytochemistry) and apoptosis (TUNEL assay) test. Furthermore, the index of Hsp27 expression and apoptosis were determined, then the data was analyzed. The results showed that IC50 of drumstick leaves extract was at 2200 μg/ml. Drumstick leaves extract was able to reduce Hsp27 expression significantly at 2200 and 4400 μg/ml (p<0.05). Drumstick leaves extract was able to increase apoptosis cells significantly at 1100, 2200, and 4400 μg/ml (p<0.05). Drumstick leaves extract as anticancer can reduce/decrease Hsp27 expression and increase apoptosis of breast cancer cell-line MCF-7.
