Abstract
Pada luka bakar derajat 2B kerap dijumpai adanya eschar tipis. Eschar merupakan jaringan mati yang melekat kuat dan memberikan risiko infeksi pada luka bakar. Pada fase inflamasi akan terjadi peningkatan makrofag karena berfungsi sebagai fagositosis. Ekstrak etanol daun cincau mengandung hidrokoloid (polisakarida pectin), antioksidan, antiinflamasi, stimulan faktor pertumbuhan (growth factor) dan imunostimulan aktivitas makrofag yang penting dalam proses penyembuhan luka bakar. Tujuan penelitian ini untuk mengetahui pengaruh ekstrak etanol daun cincau hijau (Cyclea barbata Miers) terhadap peningkatan jumlah makrofag pada fase inflamasi luka bakar derajat 2B tikus putih (Rattus novercigus Strain Wistar). Desain penelitian menggunakan true experimental dengan sampel 24 ekor tikus putih jantan yang dipilih secara simple random sampling menjadi 6 kelompok yaitu kelompok kontrol (NaCl 0,9%), kelompok perlakuan Silver Sulfadizin, kelompok perlakuan hidrogel, kelompok perlakuan ekstrak etanol daun cincau hijau konsentrasi 40%, 50%, dan 60%. Penelitian dilakukan selama 5 hari kemudian sediaan histologi jaringan kulit dibuat dengan pewarnaan Immunohistokimia (IHK). Jumlah makrofag dihitung menggunakan OlyVIA software dengan perbesaran 400x dalam 10 lapang pandang. Uji One Way ANOVA menunjukkan p-value < 0,05. Analisa Post hoc tukey HSD menunjukkan kelompok perlakuan ekstrak etanol daun cincau hijau konsentrasi 40% memiliki efek paling efektif terhadap penyembuhan luka dengan p-value 0,043. Kesimpulan yang diperoleh ekstrak etanol daun cincau hijau (Cyclea barbata Miers) dapat meningkatkan jumlah makrofag luka bakar derajat 2B pada tikus putih (Rattus novergicus strain Wistar).
English Abstract
In second degree of burn (2B-degree) is often found the presence of a thin eschar. Eschar is a necrotic tissue and provide big risk of infection in burn. During the inflammatory phase macrophage will increase because of their function as phagocytosis cell. Ethanol extract of grass jelly leaf contain hydrocolloids (polysaccharides pectin), antioxidant, anti-inflammatory, growth factor stimulation, and macrophage activity immunostimulant factor which are important in healing process of the burn. The aim of this study is to determine the effect of ethanol extract from green grass jelly leaf (Cycle barbata Miers) to increase the number of macrophages in 2B-degree burn of rats (Rattus novergicus Wistar strain). True experimental laboratory post-test only control group design was applied using 24 samples of male white rats, selected by simple random sampling into 6 groups: control group (NaCl 0.9 %), Silver Sulfadizin treatment group, hydrogel treatment group, the treatment group of ethanol extract from green grass jelly leaf with concentration 40%, 50%, and 60%. Histological preparations from skin were made by immunohistochemistry staining (IHC) in 5 days. The number of macrophages were calculated by using OlyVIA software with magnification 400x in 10 field of views. One Way ANOVA test shows p value < 0.05. Post hoc Tukey HSD analysis shows the treatment group of ethanol extract from green grass jelly leaf with concentration 40% has the most effective effect in wound healing with p value 0.043. The conclusion is an ethanol extract of green grass jelly leaf (Cyclea barbata Miers) can increase the number of macrophages in 2B degree burn of rats (Rattus novergicus Strain Wistar).
