Abstract
Antigen 38 kDa Mycobacterium tuberculosis (Ag38 kDa) memiliki spesifitas dan sensitifitas tinggi pada infeksi M. tuberculosis dan merupakan kandidat utama untuk pengembangan agen serodiagnosis untuk tuberkulosis. Produksi protein Ag38 rekombinan dilakukan dengan cara mengekspresikan gen pab pada Eshericia coli/BL21(DE3). Ag38 M. tuberculosis berhasil diekspresikan, namun produksinya masih rendah. Pada penelitian ini dilakukan upaya untuk meningkatkan produksi Ag38 rekombinan dengan cara mengkombinasikan media (LB dan 2XYT) dengan temperatur yang berbeda (37°C dan 23°C). Produksi protein dilihat melalui SDS-PAGE. Media 2XYT dan temperatur 23°C memiliki efek yang lebih baik dalam meningkatkan produksi protein Ag38 kDa dibandingkan dengan media LB dan temperatur 37°C.
English Abstract
Mycobacterium tuberculosis 38 kDa antigen (Ag38 kDa) has a high specificity and sensitivity in M. tuberculosis infection and a prime candidate for agent dvelopment in tuberculosis serodiagnosis. Production of recombinant Ag38 recombinant protein is done by expressing pab gene in Eshericia coli/BL21(DE3). M. tuberculosis Ag38 is successfully expressed, but production is still low. In this research, we improve the production of recombinant Ag38 by combining media (LB and 2XYT) at different temperatures (37°C and 23°C). Protein production is seen by SDS-PAGE. Media 2XYT and temperature 23° C has a better effect in increasing the production of Ag38 kDa protein compared with LB medium and temperature of 37°C.
