Abstract

Klebsiella pneumoniae merupakan bakteri yang paling sering menyebabkan infeksi nosokomial dan resisten terhadap beberapa obat antibiotika, oleh karena itu perlu dilakukan penelitian untuk mendapatkan antimikroba alternatif. Salah satunya batang kayu secang yang memiliki kandungan flavonoid, terpenoid, dan tanin. Penelitian ini bertujuan untuk mengetahui ekstrak batang kayu secang sebagai antimikroba terhadap Klebsiella pneumoniae. Metode yang digunakan adalah dilusi tabung yang terdiri dari tahap penentuan Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM). Ekstrak batang Kayu Secang dibuat dengan ekstraksi maserasi menggunakan etanol 96%. Konsentrasi ekstrak yang digunakan yaitu 17,5%, 16,25%, 15%, 13,75% dan 12,5%. Hasil statistik one-way ANOVA menunjukkan terdapat perbedaan yang signifikan pada perubahan konsentrasi ekstrak etanol 96% batang kayu secang terhadap jumlah koloni Klebsiella pneumoniae (p=0,000).Pada pengujian berganda (Multiple Comparasions) terdapat perbedaan jumlah koloni yang bermakna antara semua kelompok jika dibandingkan satu per satu terhadap konsentrasi 0% (p<0.05). Namun tidak terdapat perbedaan jumlah koloni yang bermakna antara konsentrasi 12,5% dengan konsentrasi 13,75% (p=0.006). Yang berarti antara dosis 12,5% dan 13,75% tidak terdapat penurunan dosis yang bermakna (p>0.05). Uji korelasi menunjukkan adanya hubungan antara konsentrasi ekstrak dengan jumlah koloni (Korelasi, R = -0,978; p=0,001) yang artinya semakin tinggi dosis / konsentrasi ekstrak etanol batang kayu secang, maka semakin rendah jumlah koloni bakteri. Berdasarkan hasil penelitian, dapat disimpulkan bahwa ekstrak batang kayu secang mempunyai efek antimikroba terhadap Klebsiella pneumoniae secara In Vitro dengan Kadar Hambat Minimum (KHM) yang tidak diketahui karena tingkat kekeruhan yang relatif sama dalam tabung dilusi dan Kadar Bunuh Minimum (KBM) adalah 17,5%.

English Abstract

Klebsiella pneumonia is bacteria that most often cause nosocomial infection and resistant to several antibiotic drugs. Therefore, it should be conducted a research to get an alternative antimicrobial. One of them is sappan wood which contains flavonoid, terpenoid, and tannin. The objective of this research is to determine sappan wood extract as antimicrobial against Klebsiella pneumoniae. The research used tube dilution method that consist of determine Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Sappan wood extract is made by maceration extraction using etanol 96%. The concentration used were 17,5%, 16,25%, 15%, 13,75% and 12,5%. The result of one-way ANOVA statistical test showed that there is significant differences in the concentration change of extract etanol 96% sappan wood on the amount of colony Klebsiella pneumoniae (p=0,000). Muliple Comparasions test showed there is significant difference between total colony in all groups if each groups was tested with 0% concentration (p<0.05). but there are no significant difference between 12,5% and 13,75% of extract concentration (p=0.006). It means that there was no significant dose decrease between 12,5% and 13,75% of extract (p>0.05). Correlation test showed that there is a correlation between concentration of extract with the amount of colony (Correlation, R = -0,978; p=0,001). It means that as increase the concentration of sappan wood stem ethanol extract will decrease of the bactery colony. Based on the result, it can be concluded that sappan wood extract has an antimicrobial effect against Klebsiella pneumoniae in vitro with the Minimum Inhibitory Concentration (MIC) which is unknown because of the relatively similar levels of turbidity in the dilution tube and Minimum Bactericidal Concentration (MBC) is 17,5%.
