Abstract

Mastitis merupakan penyakit infeksi bakteri yang disebabkan oleh spesies Staphylococcus, terutama Staphylococcus aureus. Staphylococcus aureussebenarnya adalah flora normal yang terdapat pada tubuh manusia, namun dapat berkembang biak tidak terkendali sehingga menjadi patogen karena terganggunya sistem imun. Daun srikaya dipilih sebagai alternatif pengobatan Staphylococcus aureuspada penelitian ini karena pada daun srikaya diduga memiliki bahan aktif antibakteri yaitu tannin, flavonoid, alkaloid, steroid dan saponin. Tujuan penelitian ini adalah untuk membuktikan bahwa ekstrak etanol daun srikaya dapat menghambat pertumbuhan Staphylococcus aureus. Metode penelitian eksperimental ini yaitu dengan menggunakan dilusi tabung. Konsentrasi ekstrak etanol daun srikaya yang digunakan dalam penelitian ini adalah 32,5%, 30%, 27,5%, 25%, 22,5%, 20%, dan 0%. Kadar Hambat Minimal (KHM) didapatkan dengan membandingkan tingkat kekeruhan pada masing-masing tabung uji. Sedangkan Kadar Bunuh Minimal (KBM) didapatkan dengan cara penanaman dengan penggoresan masing-masing bakteri uji pada NAP (Nutrient Agar Plate). Hasilnya didapatkan bahwa KHM adalah pada konsentrasi 30%. Sedangkan KBM pada konsentrasi 32,5%. Setelah dilakukan uji ANOVA satu arah didapatkan perbedaan bermakna antara masing masing konsentrasi ekstrak etanol daun srikaya. Dari uji korelasi Pearson didapatkan adanya hubungan yang erat antara variabel, yaitu peningkatan konsentrasi ekstrak etanol daun srikaya akan mengakibatkan penurunan jumlah koloni Staphylococcus aureus(r = -0,919). Kesimpulan dari penelitian ini yaitu ekstrak etanol daun srikaya mempunyai efek antibakteri terhadap Staphylococcus aureussecara In Vitro.

English Abstract

Mastitis is a bacterial disease caused by Staphylococcus, especially Staphylococcus aureus. Staphylococcus aureusactually normal flora that found on the human body. But it can multiply to become patogen due to disruption of the immune system. The leaves of custard apple selected as an alternative Staphylococcus aureustreatment because it has antibacterial active ingredient such as tannins, flavonoids, alkaloids, saponins, strerol and steroid. This study has purpose to prove that ethanol extract of leaves of custard apple may inhibit Staphylococcus aureus growth. This experimental research method was carried out by using tube dilution test. The concentration of ethanol extract of custard apple leaves used in this study was 32,5%, 30%, 27,5%, 25%, 22,5%, 20%, and 0%. The Minimum Inhibitory Concentration (MIC) obtained by comparing the level of turbidity in each test tube. While the Minimum Bactericide Concentration (MBC) obtained by inoculating with streaking of each test on Nutrient Agar Plates. The results found that the MIC was 30% and bactericidal concentration was 32,5%. One-way ANOVA test found the significant differences between each concentration of ethanol extract of leaves of custard apple. From the Pearson Correlation test obtained the correlation between each variable that the increasing of leaves custard apple concentration could reduce the Staphylococcus aureuscolony (r = -0,919). It was concluded that ethanol extract of custard apple leaves have antibacterial effect against Staphylococcus aureusin vitro.
