Abstract

Sindroma metabolik adalah penyakit yang terdiri dari berbagai kelainan, yaitu obesitas, aterosklerosis, resistensi insulin, protrombotik, dan hipertensi. Patogenesis sindroma metabolik karena penumpukan jaringan adiposa dalam tubuh. Hiperplasia dan hipertrofi sel adiposit pada obesitas menyebabkan peningkatan produksi leptin, TNF-α, IL-6, resistin, dan protein lain, serta penurunan produksi adiponektin sehingga mengakibatkan timbulnya sindroma metabolik. Pada kondisi sindroma metabolik jumlah Ox-LDL meningkat dan adiponektin menurun. Adiponektin adalah suatu protein yang dihasilkan adiposa dan berefek protektif terhadap sindroma metabolik. Adiponektin berperan dalam perkembangan sindroma metabolik dan menjadi penanda untuk sindroma ini. Bakteri S. Typhimurium memilki kemiripan struktur dengan Ox-LDL yang memiliki struktur Phosphorylcholine (PC) pada struktur luarnya sehingga pemberian vaksin S. Typhimurium diharapkan dapat menginduksi antibodi protektif yang bereaksi silang dengan Ox-LDL. Penelitian ini menggunakan desain eksperimen murni dan bertujuan untuk mengetahui pengaruh heat killed S. Thypimurium terhadap kadar adiponektin. Terdapat 5 kelompok perlakuan, kontrol negatif, kontrol positif, kelompok dengan diet aterogenik dan diinjeksikan vaksin berisi bakteri, kelompok dengan diet aterogenik dan diinjeksikan vaksin kombinasi ajuvan (CFA dan IFA) dan bakteri, dan kelompok dengan diet aterogenik dan diberikan vaksin ajuvan (CFA dan IFA) saja. Kadar adiponektin dalam serum tikus diukur dengan metode ELISA. Hasil dari penelitian ini menunjukkan bahwa kadar adiponektin meningkat pada kelompok dengan vaksinasi dibandingkan kontrol (+) dan kontrol (-). Sehingga dapat disimpulkan bahwa pemberian vaksin menggunakan heat killed Salmonella Typhimurium mampu meningkatkan kadar adiponektin pada tikus dengan diet aterogenik sehingga berpotensi untuk mencegah timbulnya sindroma metabolik.

English Abstract

Metabolic syndrome is a disease that consists of a variety of disorders, that is obesity, atherosclerosis, insulin resistance, prothrombotic, and hypertension. The pathogenesis of the metabolic syndrome due to a buildup of adipose tissue in the body. Cell hyperplasia and hypertrophy of adipocytes in obesity leads to increased production of leptin, TNF-α, IL-6, resistin, and other proteins, as well as decreased production of adiponectin, resulting in the onset of metabolic syndrome. In the metabolic syndrome increased Ox-LDL and decreased adiponectin happened. Adiponectin is a protein produced by adipose and has a protective effect against metabolic syndrome. Adiponectin plays a role in the development of metabolic syndrome and can be a marker for this syndrome. S. Typhimurium bacteria have the structural similarity with Ox-LDL that have phosphorylcholine (PC) structure in the outer, so vaccine with S. Typhimurium is expected to induce forming of protective antibodies that will cross react with Ox-LDL. This study uses a pure experimental design and aims to determine the effect of heat killed S. Thypimurium on levels of adiponectin. There are 5 treatment groups, negative control, positive control, a group of atherogenic diet and injected with the vaccine containing the bacteria, group atherogenic diet and injected with a combination vaccine adjuvant (CFA and IFA) and bacteria, and the group with atherogenic diet and given a vaccine adjuvant (CFA and IFA) only. Serum adiponectin levels in mice were measured by ELISA. The results of this study showed that adiponectin levels increased in the vaccinated group compared to the control (+) and control (-). It can be concluded that the vaccine using heat killed Salmonella Typhimurium is able to increase adiponectin levels in mice with atherogenic diet, and thus potentially to prevent metabolic syndrome.
