Abstract

Endocrine Disrupting Chemical adalah bahan eksogen yang mengganggu kerja regulatorik normal hormon-hormon, yang mengontrol proses perkembangan dan merupakan senyawa kimia yang mampu berikatan dengan reseptor estrogen Genistein merupakan senyawa alami yang termasuk Phytoestrogen. Salah satu jenis antagonis reseptor estrogen adalah ICI 182 780. Zebrafish (Danio rerio) sering digunakan sebagai model penelitian toksikologi, skrining obat dan organogenesis karena memiliki proses embriogenesis relatif singkat, embrio transparan dan memiliki sistem syaraf seperti mamalia. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian genistein dengan dosis tertentu (dose-dependent) terhadap respon taktil embrio zebrafish serta mekanisme kerja penghambat reseptor estrogen (estrogen blocker). Studi eksperimental ini menggunakan post test only controlled group design. Penelitian pendahuluan terdiri dari kelompok kontrol (DMSO), genistein 2,5 µM dan 5 µM. Pada penelitian selanjutnya dilakukan perbandingan paparan ICI, genistein 5 µM dan genistein 5 µM+ICI. Variabel yang diukur adalah respon taktil dengan mengambil 50 ekor pada masing-masing perlakuan dosis dan pengulangan 3 kali. Data statistik yang didapatkan adalah kelompok kontrol 98 ± 2 %, kelompok genistein 2,5 µM : 91,435 ± 1,252 %; genistein 5µM: 59,051 ± 3,51 %; ICI : 85,70213 ± 2,065 %; genistein 5 µM+ICI : 82 ± 2 %. Hasil uji Post Hoc Tukey dosis genistein 2,5 µM dan 5µM meurunkan motorik embrio zebrafish secara signifikan terhadap kontrol (p=0,035 dan p=0,000) dengan penurunan respon taktil pada larva zebrafish. Berdasarkan hasil penelitian ini, disimpulkan bahwa adanya korelasi yang signifikan (p<0,000)antara peningkatan. Pemberian ICI sebagai antiestrogen yang bekerja langsung berikatan dengan reseptor estrogen mampu berkompetisi dengan genistein sehingga mengembalikan respon taktil mendekati normal

English Abstract

Endocrine Disrupting Chemical is exogen substance which disturbs a normal regulative works of hormones controlling a development process and it is a chemical compound which is able to bond with the estrogen receptor. Genistein is a natural compound included Phytoestrogen. One kind of the antagonist receptor of estrogen is ICI 182 780. Zebrafish (Danio rerio) is often used as a model for toxicology research, medicine screening and organogenesis because they have an relatively short empriogenesis process, transparent embrio and having nerve system, like mamalia. This research aims to know the influence of giving genistein in a certain dose (dose-dependent) towards tactile response of zebrafish embrio , and the mechanism of the estrogen blocker. The experimental study uses a post test only controlled group design. The first research consists of controlled group (DMSO), genistein 2,5 µM and 5 µM. Then, the next research is carried out comparative ICI, genistein 5 µM and genistein 5 µM+ICI. The measured variabel is tactile response by taking 50 fish for each dose treatment and three times of repetition. Then, the found statistics data is the controlled group 98 ± 2 %, the genistein group 2,5 µM : 91,435 ± 1,252 %; genistein 5µM: 59,051 ± 3,51 %; ICI : 85,70213 ± 2,065 %; genistein 5 µM+ICI : 82 ± 2%. Based on those research, it is concluded that there is a significant ( p<0,000) correlation between the increasing dose of genistein and the decreasing of tactile response in zebrafish larva. Giving ICI as antiestrogen which directly works bonded in estrogen receptor is able to compete with genistein , so it will return the tactile response approaching to normal.
