Abstract
Karies merupakan masalah yang sering dijumpai dalam praktek kedokteran gigi. Salah satu penyebabnya adalah Streptococcus mutans yang merupakan bakteri flora normal pada rongga mulut. Daun meniran mempunyai bahan aktif yang diduga memiliki efek antibakteri, yaitu flavonoid, tannin, dan alkaloid. Penelitian ini bertujuan untuk membuktikan efek antibakteri daun meniran (Phyllanthus niuri Linn) terhadap Streptococcus mutans. Desain penelitian yang digunakan adalah eksperimental laboratoris (true experiment–post test only control group design) dengan menggunakan metode dilusi agar dan dilusi tabung. Ekstrak daun meniran diperoleh dengan cara ekstraksi dengan pelarut etanol 96% menggunakan metode maserasi. Konsentrasi ekstrak dau meniran yang digunakan adalah 12,5%, 15%, 17,5%, 20%, 22,5%, dan 25%. Jumlah isolat Streptococcus mutans adalah satu isolat dengan empat kali pengulangan. Kepadatan bakteri yang diuji adalah 10⁶ CFU/ml. Dari hasil penelitian, didapatkan Kadar Hambat Minimum (KHM) ekstrak daun meniran terhadap Streptococcus mutans sebesar 15%. Sedangkan Kadar Bunuh Minimum (KBM) pada konsentrasi 25%. Hasil analisis statistik menunjukkan bahwa ekstrak daun meniran secara signifikan dapat menghambat pertumbuhan Streptococcus mutans secara in vitro (ANOVA, p<0,005) dan terdapat pengaruh yang kuat antara konsentrasi ekstrak daun meniran dengan pertumbuhan Streptococcus mutans (korelasi R=-0,894). Kesimpulan yang didapat adalah ekstrak daun meniran memiliki efek antibakteri terhadap Streptococcus mutans secara in vitro. Diperlukan penelitian lebih lanjut dengan memperbanyak sampel peelitian atau pengulangan. Selain itu perlu diteliti efek daun meniran baik terhadap hewan coba maupun manusia.
English Abstract
Caries is a frequent problem encountered in tepractice of dentistry. One possible cause is Streptococcus mutans. Phyllanthus ninuri Linn has active ingredients that are thought to have antibacterial effects, namely flavonoid, tannin, and alkaloid. The aim of the study to prove the antibacterial effect of Phyllanthus ninuri Linn against Streptococcus mutans. Design research is an experimental laboratory (true experiment - post test only control group design) by using the method of dilution agar and dilution tube. Phyllanthus ninuri Linn extract was purified using 96% ethanol with maceration method. Phyllanthus ninuri Linn extract used was 12,5%, 15%, 17,5%, 20%, 22,5%, and 25%. The number of isolates of Streptococcus mutans is one isolate with four repetitions. The densityof the bacteria tested is 10⁶ CFU/ml. From the research result, the MIC of Phyllanthus ninuri Linn extract against Streptococcus mutans is at 25%. While Minumum Bactericidal Concentration (MBC) at 25%. The results of statistical analysis showed that Phyllanthus ninuri Linn extract can significantly inhibit the growth of Streptococcus mutans in vitro (ANOVA, p<0.05) and there is a strong influence between the concentrations of Phyllanthus ninuri Linn extract with Streptococcus mutans colony growth (corelation R = -0,894 ). The result is Phyllanthus ninuri Linn extract have shown to have antibacterial effect againts Streptococcus mutans. Further research is needed to reproduce the study sample. Ultimately, the effect of Phyllanthus ninuri Linn needs to be tested both on animal and human.
