Abstract
Penyakit kardiovaskuler merupakan salah satu penyakit yang banyak diderita di dunia dengan tingkat mortalitas dan morbiditas yang tinggi. Penyebab utama dari penyakit kardiovaskuler adalah aterosklerosis. Pada penderita penyakit kardiovaskuler terjadi perkembangan lesi aterosklerosis pada dinding pembuluh darahnya. Aterosklerosis merupakan penyakit progresif yang menginduksi inflamasi kronis yang menyebabkan stenosis, oklusi, dan penimbunan material-material proaterogenik pada arteri besar sehingga menyebabkan penebalan pada dinding aorta. Salah satu strategi terbaru untuk mencegah aterosklerosis adalah dengan menggunakan antioksidan. Daun bayam (Amaranthus hybidus) memilki kandungan antioksidan yang cukup tinggi, diantaranya β-karoten, vitamin C, vitamin E, flavonoid, dan saponin. Pemberian ekstrak daun bayam pada tikus terbukti menurunkan respon inflamasi tubuh. Tikus dibagi menjadi 3 kelompok perlakuan, kontrol positif, dan kontrol negatif. Induksi aterosklerosis menggunakan diet aterogenik. Pengukuran lesi aterosklerosis dilakukan dengan melihat histopatologi aorta tikus kemudian diukur ketebalannya. Pemberian diet aterogenik secara signifikan (p < 0,05) meningkatkan ketebalan aorta tikus. Pemberian ekstrak daun bayam dosis 600 mg/kgBB pada tikus yang diberi diet aterogenik secara signifikan (p < 0,05) mampu mencegah peningkatan ketebalan aorta. Hasil uji statistik One Way ANOVA terhadap data menunjukkan adanya perbedaan yang signifikan antar kelompok, yaitu p = 0, 000 (p < 0,05), sementara hasil uji korelasi menunjukkan hubungan yang kuat antara dosis ekstrak metanol daun bayam dengan ketebalan aorta tikus (R = -0,859; p = 0,000). Kesimpulan dari penelitian ini adalah pemberian ekstrak metanol daun bayam mampu mencegah peningkatan ketebalan aorta tikus Wistar yang diberi diet aterogenik secara signifikan, dengan hasil maksimal pada dosis 600 mg/kgBB.
English Abstract
Cardiovascular disease is one disease that affects many in the world with a high rate of mortality and morbidity. The main cause of cardiovascular disease is atherosclerosis. In patients with cardiovascular disease, atherosclerotic lesion development occurred on the walls of blood vessels. Lesions of atherosclerosis is a progressive disease that causes chronic inflammation induced stenosis, occlusion, and stockpiling materials proaterogenik in large arteries, causing thickening of the aortic wall. One of the latest strategies to prevent atherosclerosis is by using antioxidants. Leaf spinach (Amaranthus hybidus) have a fairly high content of antioxidants, including β - carotene, vitamin C, vitamin E, flavonoid, and saponin. Spinach leaf extract in mice shown to decrease the bodys inflammatory response. Rats were divided into 3 treatment groups, the positive control, and negative control. Induction of atherosclerosis using atherogenic diet. Measurement of atherosclerotic lesions is done by looking at the histopathology rat aorta is then measured its thickness. Giving atherogenic diet significantly (p <0.05) increases the thickness of the rat aorta. Spinach leaf extract dose of 600 mg/kgBW in mice fed atherogenic diet significantly (p <0.05) were able to prevent an increase in the thickness of the aortic. Results of One Way ANOVA statistical test on the data showed significant differences between groups, ie p = 0 , 000 ( p < 0.05 ), while the correlation test results demonstrate a strong relationship between the dose of methanol extract of leaves of spinach with a thickness of rat aorta (R = -0.859, p = 0.000). The conclusion of this study is the methanol extract of leaves of spinach were able to prevent an increase in the thickness of the aortic Wistar rats fed atherogenic diet significantly, with maximum results at a dose of 600 mg/kgBW
