Abstract

Salmonella typhi adalah bakteri penyebab demam tifoid pada anak maupun orang dewasa. Salmonella typhi ditularkan melalui makanan dan minuman yang terkontaminasi. Selama beberapa dekade terakhir, resistensi antibiotik terhadap Salmonella typhi semakin meningkat sehingga menimbulkan masalah terapi yang sulit. Salah satu alternatif terapi adalah dengan bahan alami, yaitu daun sukun (Artocarpus altilis). Kandungan aktif pada daun sukun yang diduga bermanfaat sebagai antimikroba adalah flavonoid, tannin, saponin dan kuinon. Penelitian ini bertujuan untuk mengetahui potensi antimikroba ekstrak etanol daun sukun (Artocarpus altilis) terhadap Salmonella typhi. Penelitian ini merupakan penelitian eksperimental laboratoris murni dengan post test only control group design, menggunakan metode dilusi tabung. Sampel yang digunakan dalam penelitian ini adalah Salmonella typhi yang diperoleh dari Balai Laboratorium Kesehatan Yogyakarta. Konsentrasi ekstrak daun sukun yang digunakan yaitu 3%, 3,5%, 4%, 4,5% dan 5% dengan empat kali pengulangan. Data yang didapat dianalisis dengan uji statistik one way anova, uji multi komparasi Pos Hoc Tukey, uji korelasi Pearson dan uji Regresi Linear dengan tingkat kepercayaan (α) = 0,05. Hasilnya menunjukkan bahwa ekstrak daun sukun (Artocarpus altilis) mempunyai efek sebagai antimikroba terhadap Salmonella typhi dapat dilihat dari Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM).

English Abstract

Salmonella typhi is the bacteria that causes typhoid fever in children and adults. Salmonella typhi is transmitted through contaminated food and drink. Over the past few decades, antibiotic resistance against Salmonella typhi is increasing causing a difficult therapeutic problem. One alternative therapy is using natural substance, i.e. breadfruit leaf (Artocarpus altilis). The active compositions of breadfruit leaf (Artocarpus altilis) which allegedly useful as antimicrobial are flavonoid, tannin, saponin and quinone. This study aims to detemine the antimicrobial potency of breadfruit leaf (Artocarpus altilis) extract against Salmonella typhi. This study is an experimental research laboratory using post test only control group design, done by tube dilution method. Samples used in this study were Salmonella typhi derived from Health Laboratory of Yogyakarta. Consentrations of the breadfruit leaf extract used are 3%, 3,5%, 4%, 4,5% and 5% with four repetitions. The data obtained were analyzed by one way anova statistical test, multiple comparison test of Post Hoc Tukey, Pearson correlation test and linear regression test with a confidence level (α) = 0.05. The results showed that the breadfruit leaf (Artocarpus altilis) extract has an antimicrobial effect against Salmonella typhi as can be seen from the Minimum Inhibitory concentration (MIC) and Minimum Bactericidal Concentration (MBC).
