Abstract
Staphylococcus aureus merupakan bakteri normal atau sering disebut flora normal dalam tubuh manusia. Faktor yang membuat bakteri ini sangat penting karena Staphylococcus aureus sangat sering menyebabkan berbagai penyakit infeksi dalam tubuh seperti keracunan makanan, infeksi kulit sampai kajadian yang membahayakan yaitu sindroma syok toksik, serta yang tidak kalah penting kaitannya dengan kejadian multiresisten terhadap beberapa antibiotik. Biji gambas (Luffa acuntangula) mengandung senyawa saponin yang ini merupakan senyawa glikosida kompleks bermolekul tinggi yang mengandung gugus gula terutama glukosa, galaktosa, xylosa, rhamnosa atau methilpentosa yang berikatan dengan suatu aglikon hidrofobik (sapogenin) berupa triterpenoid atau steroid alkaloid yang berfungsi sebagai antimikroba melalui proses penghambatan sintesis dinding sel bakteri serta proses denaturasi protein. Penelitian ini bertujuan untuk mengetahui efek antimikroba ekstrak biji gambas (Luffa acuntangula) pada pertumbuhan bakteri Staphylococcus aureus in vitro. Desain penelitian ini adalah desain eksperimental laboratorium. Ekstrak etanol menggunakan metode maserasi dengan pelarut etanol 96%. Metode dilusi tabung digunakan untuk mengetahui Kadar Hambat Minimal (KHM) dan Kadar Bunuh Minimal (KBM). Konsentrasi ekstrak yang digunakan yaitu 0% (kontrol positif), 25 %, 30 %, 35 %, 40 %, 45 %, 50 % dan 55 %. Pada penelitian ini KHM tidak dapat ditemukan dan KBM berada pada konsentrasi ekstrak 50%. Hasil penelitian menunjukkan secara signifikan bahwa peningkatan konsentrasi ekstrak biji gambas (Luffa acuntangula) menyebabkan penurunan tingkat pertumbuhan bakteri Staphylococcus aureus. Kesimpulan dari penelitian ini adalah ekstrak biji gambas (Luffa acuntangula) memiliki efek antimikroba terhadap pertumbuhan Staphylococcus aureus in vitro.
English Abstract
Staphylococcus aureus is a normal bacterium or often called the normal flora in the human body . The factor that makes this bacterium Staphylococcus aureus very important is because Staphylococcus aureus often lead to a variety of infectious diseases in the human body such as food poisoning , skin infection until the most dangerous effect called toxic shock syndrome , also other important effect is the relation of Staphylococcus aureus to the multiresistant antibiotic incidence. Gambas seeds (Luffa acuntangula) contains saponin glycosides thats a complex compound, contains high biomolecule group containing sugar mainly glucose, galactose, xylose, or methilpentosa rhamnosa that binds to a hydrophobic aglycone (sapogenin) a triterpenoid or steroidal alkaloid that functions as an antimicrobial through the inhibition of bacterial cell wall synthesis and protein denaturation process. This study aims to determine the antimicrobial effect of gambas seed extract (Luffa acuntangula ) on the growth of Staphylococcus aureus in vitro . The design of this study is the design of experimental laboratory . The ethanol extract using maceration method with 96 % ethanol . Tube dilution method is used to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). The concentration of extract used is 0 % ( positive control) , 25 % , 30 % , 35 % , 40 % , 45 % , 50 % and 55 % . In this study the MIC could not be found and MBC to be at 50 % concentration of the extract . The results showed that by increasing concentration of gambas seeds extract ( Luffa acuntangula ) causes a significant decrease in the growth rate of the Staphylococcus aureus . The conclusion of this study is the gambas seeds extract ( Luffa acuntangula ) has an antimicrobial effect on the growth of Staphylococcus aureus in vitro .
