Abstract

Shigella dysenteriae adalah bakteri gram negatif dari famili Enterobacteriaceae yang merupakan penyebab utama disentri basiler di negara berkembang dengan insiden multi drug resistance yang tinggi. Daun pecut kuda ( stachytarpheta jamaicensis L. ( vahl )) diketahui memiliki bahan-bahan aktif yaitu alkaloid, flavonoid, saponin, tanin yang mempunyai efek antimikroba. Penelitian ini bertujuan untuk mengetahui dosis ekstrak etanol daun pecut kuda yang paling efektif sebagai antimikroba terhadap Shigella dysenteriae secara in vitro . Penelitian ini merupakan penelitian eksperimental yang menggunakan the post test only control group design . Untuk menentukan KHM (Kadar Hambat Minimal) digunakan metode dilusi agar sedangkan untuk menentukan KBM (Kadar Bunuh Minimal) digunakan metode dilusi tabung. Konsentrasi ekstrak yang digunakan adalah 0%, 15%, 16%, 17%, 18%, dan 19% dengan empat kali pengulangan. Hasil penelitian ini didapatkan KHM sebesar 17% dan KBM sebesar 19%. Analisis data statistik menunjukkan adanya penurunan jumlah koloni bakteri Shigella dysenteriae yang signifikan seiring dengan peningkatan dosis ekstrak etanol daun pecut kuda ( Stachytarpheta jamaicensis L. (Vahl)) (Pearson, p<0,05). Berdasarkan penelitian ini dapat disimpulkan bahwa ekstrak etanol daun pecut kuda efektif sebagai antimikroba terhadap Shigella dysenteriae secara in vitro.

English Abstract

Shigella dysenteriae is a gram negative bacteria of the Enterobacteriaceae family is a major cause of bacillary dysentery in the developing countries with high incidence of multi drug resistance. Blue potterweed leaves ( Stachytarpheta jamaicensis L. (Vahl)) is known to have active substances such as alkaloids, flavonoids, saponins, and tannins that have antimicrobial effect. This study aims to determine the most effective antimicrobial dose of the blue potterweed leaves against Shigella dysenteriae in vitro . This research is an experimental study using the the post test only control group design. Agar dilution method was used to determine the MIC value while tube dilution method was used to determine the MBC value. Concentration of the extract used were 0%, 15%, 16%, 17%, 18%, and 19% with four repetitions. The result of this study was 17% for MIC and 18% for MBC. Statistical analysis of the data shows there is a significant decrease on the amount of Shigella dysenteriae colonies with the increasing doses of blue potterweed leaves ( Stachytarpheta jamaicensis L. (Vahl)) ethanol extract (Pearson, p<0,05). Based on this study it can be concluded that the ethanol extract of the blue potterweed leaves ( Stachytarpheta jamaicensis L. (Vahl )) is effective as an antimicrobial against Shigella dysenteriae in vitro.
