Abstract

Diabetes Mellitus (DM) tipe 2 akibat penurunan sensitivitas insulin merupakan penyebab 90-95% kasus DM di dunia. Peningkatan ekspresi regulatory subunit enzyme p85 dari phosphatidylinositol (PI) 3-kinase dipercaya bertanggung jawab terhadap terjadinya resistensi insulin pada penderita DM tipe 2. Studi imunologis menunjukkan penggunaan p85 sebagai antigen mampu menginduksi pembentukan antibodi yang mampu berikatan secara spesifik dengan enzim tersebut. Tujuan penelitian ini adalah mengetahui potensi p85 dalam menginduksi antibodi protektif terhadap DM tipe 2 sebagai vaksinasi untuk pencegahan DM tipe 2. Penelitian ini adalah eksperimen murni dengan metode Randomized Post Test Only Controlled Group Design . 25 tikus wistar jantan usia 8 minggu dibagi menjadi 5 kelompok yaitu kontrol positif, kontrol negatif, p85-CFA/IFA 100μl, p85 100μl, CFA/IFA 100μl dan dinduksi DM tipe 2 dengan diet induced obesity selama 4 minggu serta streptozotocin (STZ) pada minggu ke-5 dengan dosis 30mg/kgbb. Injeksi vaksin diberikan 2 minggu sekali sebanyak 3 kali secara intraperitoneal. Kadar glukosa darah dimonitoring tiap 10 hari sekali mulai hari ke-0. Tikus dibedah hari ke-37 dan diambil sampel darah serta jaringan lemaknya. Pengukuran kadar p85 pada jaringan lemak menggunakan ELISA. Pada penelitian ini pemberian vaksinasi p85 mampu menurunkan kadar p85 pada jaringan lemak tikus dan berat lemak perut pada tikus yang diinduksi DM tipe 2 secara signifikan (p<0,05). Selain itu, terdapat kecenderungan penurunan kadar glukosa darah pada tikus yang diberi vaksinasi. Berdasarkan hasil tersebut dapat disimpulkan bahwa vaksinasi dengan p85 mampu menginduksi pembentukan antibodi yang bersifat protektif terhadap DM tipe 2 secara in vivo.

English Abstract

90-95% cases of Diabetes Mellitus in the world are Type 2 Diabetes Mellitus (DM) that caused by the insulin sensitivity depression The enhancement of regulatory subunit enzyme p85 expression of phosphatidylinositol (PI) 3-kinase is believed to be responsible for the development of insulin resistance in patients with type 2 diabetes mellitus. Immunological studies showed that p85 usage as an antigens were able to induce the formation of antibodies that can bind specifically to the enzyme. The aim of this study is to know potency of p85 in inducing type 2 DM protective antibody formation as a vaccine for type 2 DM prevention. This was a purely experimental study using the method of Randomized Post Test Only Controlled Group Design. Twenty five male wistar rats aged of 8 weeks was divided into 5 groups: positive control, negative control, p85-CFA/IFA 100μl, 100μl P85, and CFA/IFA 100μl. Rats were induced as type 2 DM by diet induced obesity for 4 weeks and the administration of streptozotocin (STZ) on week 5 with the dosage of 30mgs per body weigh. The vaccine was given 3 times intraperitoneally, with the duration of once in 2 weeks. Blood glucose levels was monitored every 10 days, started from day 0. Rat was operated in the day of 37 to take its blood and fat tissue as sample. P85 expression in fat tissue was measured by using ELISA. In this study, vaccination can lowering P85 levels in rat adipose tissue and abdominal fat weight in rat induced type 2 diabetes mellitus significantly (p<0,05). In addition, there is a tendency to lowering blood glucose levels in rat given vaccination. Based on these results it can be concluded that vaccination with P85 is able to induce the formation of antibodies that are protective against type 2 diabetes in vivo.
