Abstract
Acinetobacter baumannii merupakan patogen opportunistik atau patogen nosokomial yang dapat menyebabkan beberapa penyakit seperti bakteremia, meningitis, pneumonia dan infeksi saluran kemih yang mengakibatkan morbiditas yang parah dan angka mortalitas yang tinggi. Oleh karena infeksi ini didapat di rumah sakit, maka bakteri ini pada umumnya resisten terhadap banyak antibiotika. Oleh karena itu perlu dikembangkan alternatif pengobatan yang lebih efektif, efisien, dan lebih murah tanpa melupakan standar mutu pelayanan medis. Kulit buah manggis diketahui mengandung bahan aktif yang bersifat antibakteri. Penelitian ini bertujuan untuk membuktikan efek antibakteri ekstrak etanol kulit buah manggis terhadap Acinetobacter baumannii secara in vitro . Metode yang digunakan adalah uji dilusi agar yang bertujuan untuk menentukan Kadar Hambat Minimum (KHM) dan uji dilusi tabung untuk menentukan Kadar Bunuh Minimum (KBM). Ekstrak kulit buah manggis dibuat dengan cara maserasi menggunakan etanol 96%. Konsentrasi ekstrak etanol kulit buah manggis yang digunakan adalah 0,3%, 0,4%, 0,5%, 0,6% dan 0,7%. Hasil uji statistik menunjukkan bahwa ekstrak etanol kulit buah manggis secara signifikan dapat menghambat pertumbuhan Acinetobacter baumannii (ANOVA, p = 0.000, p<0,05) dan terdapat hubungan antara peningkatan konsentrasi ekstrak etanol kulit buah manggis dengan penurunan jumlah pertumbuhan koloni Acinetobacter baumannii (R = -0.823). KHM ekstrak etanol kulit buah manggis terhadap Acinetobacter baumannii adalah 0,5% dan KBMnya adalah 0,7%. Kandungan ekstrak etanol kulit buah manggis yang diduga berperan sebagai antibakteri adalah xanton, saponin, tanin, terpenoid dan flavonoid. Sehingga dapat disimpulkan bahwa ekstrak etanol kulit buah manggis memiliki efek antibakteri terhadap Acinetobacter baumannii secara in vitro .
English Abstract
Acinetobacter baumannii is an opportunistic pathogen or nosocomial pathogen that can cause several diseases like bacteremia, meningitis, pneumonia, and urinary tract infection that lead to severe morbidity and high mortality rate. This infection is obtained at hospital, causing this bacteria resistant against antibiotics Therefore an effective alternative medication should be developed. Mangosteen peel is known contains active materials that can be used as antibacteria. This experiment aims to verify antibacterial effect of ethanol extract mangosteen peel against Acinetobacter baumannii in vitro. This experiment using agar dilution method to determine Minimum Inhibitory Concentration (MIC) and tube dilution method to determine Minimum Bactericide Concentration (MBC). Mangosteen peel extract was made by maceration with 96% ethanol. The concentration of mangosteen peel extract were 0,3%, 0,4%, 0,5%, 0,6% and 0,7%. Statistical analysis showed that ethanol extract of mangosteen peel could significantly inhibit growth of Acinetobacter baumanii (ANOVA, p=0.000, p<0,05) and there is a correlation between the increase of ethanol extract of mangosteen peel concentration with the decrease of colonies growth of Acinetobacter baumannii (R = -0.823). This experiment showed that MIC of ethanol extract of mangosteen peel to Acinetobacter baumannii was 0,5% and MBC was 0,7%. The active matherials of ethanol extract of mangosteen peel that expected as antibacteria are xanton, saponin, tanin, terpenoid and flavonoid. It was concluded that ethanol extract of mangosteen peel has an atibacterial effect against Acinetobacter baumannii in vitro.
