Abstract
Bibir sumbing disebabkan oleh banyak faktor dan salah satunya adalah genetik. EGFR adalah salah satu growth factor yang mengaktifkan proliferasi dan ERK-1 merupakan salah satu jalur dari MAPK yang jalurnya diaktifkan oleh EGFR. Sampai saat ini masih belum ada penelitian yang menjelaskan apakah ada ekspresi dari protein EGFR dan ERK-1 pada kejadian bibir sumbing. Penelitian ini ingin mengetahui bagaimana hubungan anatara protein EGFR dan ERK-1 pada kejadian bibir sumbing ras protomalayid di Provinsi Nusa Tenggara Timur. Penelitian ini menggunakan desain penelitian cross sectional analytic . Sampel penelitian adalah jaringan bibir sumbing hasil operasi oleh tim bedah plastik rumah sakit Saiful Anwar Malang pada kegiatan bakti sosial tanggal 3,6,7,8 dan 12 Desember 2012 Di RSU Larantuka Kupang, RSU Kupang, RSU Alor Nusa Tenggara Timur yang memenuhi kriteria inklusi eksklusi. Jaringan bibir dilakukan pewarnaan imunohistokimia masingmasing jaringan dengan antibody monoklonal EGFR dan antibodi monoklonal ERK-1 setelah itu hasil pewarnaan tersebut akan di scan dan dihitung menggunakan program OlyVia. Analisis korelasi antara protein EGFR dan ERK-1 diuji dengan uji Pearson. Hasil penelitian menunjukan hubungan yang bermakna dari ekspresi protein EGFR dan ekspresi protein ERK-1 dengan koefisien korelasi yang cukup (0.369). Kesimpulan dari penelitian ini adalah terdapat hubungan yang positif dan bermakna dengan koefisien korelasi yang cukup kuat antara ekspresi protein EGFR dan ekspresi protein ERK-1 pada kejadian bibir sumbing ras Protomalayid provinsi Nusa Tenggara Timur.
English Abstract
Cleft lip is caused by many facotrs and one of those factors is genetic. EGFR is one of growth factor that activating proliferation process and ERK-1 is one of MAPK pathway which is activated by EGFR. This research This research is conducted to understand the correlation between EGFR protein and ERK-1 protein on Protomalayid people with cleft lip in East Nusa Tenggara. The design of this research is cross sectional analytic. Sample of this research is cleft lip tissue acquired by Saiful Anwar Hospital’s plastic surgeon team taken during social event which held on 3,6,7,8 and 12 December 2012 at Larantuka Hospital of Kupang, Hospital of Kupang, Hospital of Alor which all located in Easr Nusa Tenggara that fulfill the exclusion and inclusion criteria. Each of the cleft lip tissue is stained with immunohistochemistry staining process using monoclonal antibody EGFR and monoclonal antibody ERK-1 then scanned and counted by OlyVia programme . Correlation analysis between EGFR and ERK-1 is done by Pearson test. The result reveals a significance correlation between EGFR and ERK-1 protein expression with moderate coefficient correlation(0,369). The conclusion is TGF-β1 and p38 MAPK have a significance correlation with moderate coefficient correlation on Protomalayid race with cleft lip in East Nusa Tenggara province.
