Abstract
Dm Tipe 2 Akibatpenurunansensitivitas Insulin Merupakanpenyebab 90-95% Kasus Dm Di Dunia.Peningkatanekspresi regulatory Subunit Enzyme P85 dari phosphatidylinositol (Pi) 3-Kinase Dipercayabertanggungjawabterhadapterjadinyaresistensi Insulin Padapenderita Dm Tipe 2.Studiimunologismenunjukkanpenggunaan P85 Sebagai Antigen Mampumenginduksipembentukanantibodi Yang Mampuberikatansecaraspesifikdenganenzimtersebut.Tujuanpenelitianiniadalah M engetahui Potensip85 Dalammenginduksiantibodiprotektifterhadap Dm Tipe 2 sebagaivaksinasiuntukpencegahan Dm Tipe 2 Dengan Advanced Glycation End Product (Age) Sebagai Markernya . Penelitianiniadalah eksperimen Murni Denganmetode randomized Post Test Only Controlled Group Design . 25 Tikuswistarjantanusia 8 Minggudibagimenjadi 5 Kelompokyaitukontrolpositif, Kontrolnegatif, P85-Cfa/Ifa 100μl , P85 100μl , Cfa/Ifa 100μl Dandinduksi dm Tipe 2 Dengan Diet Induced Obesity Selama 4 Minggu serta streptozotocin (Stz) Padaminggu Ke-5 Dengandosis 3 0mg/Kgbb. Injeksivaksindiberikan 2 Minggusekalisebanyak 3 Kali Secaraintraperitoneal. Kadar Glukosadarahdimonitoringtiap 10 Harisekalimulaihari Ke-0. Tikusdibedahharike- 37 dandiambilsampeldarahnya. Pengukuran Kadar Agepada Serum Dilakukan dengan Elisa. Padapenelitianinipemberianvaksinasi P85 Mampumenurunkan kadar Age Pada Tikus Yang Diinduksi Dm Tipe 2 Secarasignifikan (P < 0,05). Berdasarkan Hasil Tersebut Dapat Disimpulkan Bahwa Vaksinasi Dengan P85 mampu Menginduksi Pembentukan Antibodi Yang Bersifat Protektif Terhadap Dm Tipe 2 Secara In Vivo.
English Abstract
ype 2 Dm (T2dm) Is The Most Common Form Of Diabetes, Accounting For 90-95% Of Cases. In T 2 dm , The Body Does Not Respond Properly To Insulin, A Condition Known As Insulin Resistance . Overexpression Of Regulatory Subunit Enzyme P85 From Phosphatidylinositol (Pi) 3-Kinase Signaling Is Thought To Play An Essential Role In T2dm Insulin Resistance. Immunological Studies Show That The Used Of P85 As An Antigen Can Induce Production Of Antibody Which Specifically Bind To That Enzyme. This Research Aim To Investigating About P85 Potential In Induce T2dm Protective Antibody So It Can Be Used As A Vaccine To Prevent This T2dm Disease With Advanced Glycation End Product (Age) As A Marker. This Research Is A True Experimental Study With Randomized Post Test Only Controlled Group Design. 25 Male Rats Aged 8weeks Was Divided Into 5 Group, They Arepositive Control, Negative Control, P85-Cfa/Ifa 100 μ l, P85 100 μ l, Cfa/Ifa 100 μ l. Inducing T2dm Was Done By Diet Induced Obesity For 4 Weeksand30mg/Kgbb Streptozotocin (Stz) Injection On Fifth Week. Vaccine Injection Was Given Intraperitoneal Three Times Every 2 Weeks.Blood Glucose Levels Were Monitored Every 10 Day Since Day 0.On 37th Day, Rats Were Sacrificed And Their Blood Was Collected As A Sample. Tests Of Ages Level On Serum Were Done With Elisa Method. The Result Shows That P85 Vaccination Significantly (P<0,05) Lower Serum Age Level In Rats Induced With T2dm. Based On This Result, We Conclude That P85 Vaccination Induce Protective Antibody To T2dm In Vivo.
