Uji Efektivitas Ekstrak Etanol Daun Mangga (Mangifera Indica L.) Terhadap Pertumbuhan Bakteri Shigella Dysenteriae Secara In-Vitro

Abstract

Shigella Dysenteriae Merupakan Bakteri Enterik Penyebab Diare Akut Pada Manusia. Saat Ini Terjadi Pola Pergeseran Resistensi Obat Antimikroba, Utamanya Di Negara Berkembang. Diperlukan Sumber Antimikroba Lainnya Untuk Menangani Masalah Tersebut Dan Salah Satunya Terdapat Pada Tanaman Herbal. Daun Mangga ( Mangifera Indica L.) Yang Memiliki Kandungan Flavonoid, Saponin, Dan Tanin, Memiliki Kemampuan Sebagai Antimikroba. Penelitian Ini Bertujuan Untuk Mengetahui Efek Ekstrak Etanol Daun Mangga Sebagai Antimikroba Terhadap Shigella Dysenteriae Secara In Vitro . Penelitian Ini Adalah True Experimental Post Test Only Design Dengan Metode Difusi Cakram Yang Terdiri Dari Tahap Penentuan Kadar Hambat Minimum (Khm) Dan Tahap Penentuan Kadar Bunuh Minimum (Kbm). Sampel Bakteri Diperoleh Dari Isolat Klinik Laboratorium Mikrobiologi Universitas Brawijaya. Daun Mangga Diekstraksi Menggunakan Metode Maserasi Dengan Pelarut Etanol 96%. Konsentrasi Ekstrak Etanol Daun Mangga Yang Dipakai Pada Penelitian Ini Yaitu 7%, 8%, 9%, 10%, 11%, Dan 12% Dengan Dua Kontrol, Kontrol Positif Dan Kontrol Negatif. Data Hasil Penelitian Dianalisa Dengan Uji One Way Anova Dan Uji Korelasi Dan Regresi Dengan Tingkat Kepercayaan 95% (Α = 0,05). Dari Hasil Uji One-Way Anova Terdapat Perbedaan Yang Signifikan (0,000) Pada Perubahan Konsentrasi Ekstrak Terhadap Jumlah Koloni Bakteri ( One-Way Anova , P < 0,05). Uji Korelasi Dan Regresi Menunjukkan Bahwa Semakin Meningkatnya Konsentrasi Ekstrak Maka Semakin Sedikit Jumlah Koloni Bakteri (R= -0.894, P < 0,05). Dari Hasil Penelitian Ini Dapat Disimpulkan Bahwa Ekstrak Etanol Daun Mangga Efektif Sebagai Antimikroba Terhadap Shigella Dysenteriae Secara In Vitro Dengan Kadar Hambat Minimum (Khm) Adalah 3,125% Dan Kadar Bunuh Minimum (Kbm) Adalah 12%. Disarankan Untuk Dilakukan Penelitian Lanjutan Dengan Uji In Vivo Dan Uji Klinik Agar Hasil Penelitian Bisa Diterapkan Dalam Pelayanan Kesehatan.

English Abstract

Shigella Dysenteriae Is An Enteric Bacteria That Cause Acute Diarrhea. Nowadays, There Is Shifting Pattern Of Antimicrobial Drug Resistence, Especially In Developing Countries. It Is Needed Another Sources Of Antimicrobial To Treat That Problem And One Of Them Can Be Found In Herbal Plants. Mango Leaves ( Mangifera Indica L. ) Have Compounds Such As Flavonoid, Saponin, And Tannin That Have Ability As Antimicrobial. The Aim Of This Study Was To Know The Antimicrobial Effect Of Ethanol Extract Of Mango Leaves To Shigella Dysenteriae In Vitro . This Research Was True Experimental Post Test Only Design With Disk Diffusion Method Consisting Of Phase Determination Of Minimum Inhibitory Concentration (Mic) And Phase Determination Of Minimum Bactericidal Concentration (Mbc). Sample Was Obtained From Clinical Isolates In The Microbiology Laboratory Of Brawijaya University. Mango Leaves Was Extracted By Maseration Method With The Solvent Was Ethanol 96%. The Concentrations Of Ethanol Extract Of Mango Leaves That Is Used In This Research Were 7%, 8%, 9%, 10%, 11%, And 12% With Two Controls, Positive Control And Negative Control. Data From This Research Was Analyzed By One Way Anova Test And Correlation And Regression Test With Confidence Interval 95% (Α = 0,05). From The One Way Anova Test, There Were Significant Differences Effect (0.000) In The Changes Of The Extract Concentrations To The Total Bacterial Colony ( One-Way Anova, P<0,05). Regression Correlation Test Showed That The Higher Extract Concentration, The Smaller Of Total Bacterial Colony (R= -0,894, P<0,05). Based On The Result, It Can Be Concluded That The Ethanol Extract Of Mango Leaves Has An Antimicrobial Effect Against Shigella Dysenteriae In Vitro With The Minimum Inhibitory Concentration (Mic) Is 3,125% (V/V) And Minimum Bactericidal Concentration (Mbc) Is 12% (V/V). It Is Recommended For Further Research With In Vivo Testing And Clinical Testing, So That The Result Can Be Applied In Health Services.
