Pengaruh Aktivitas Antibakteri Ekstrak Daun Juwet (Syzygium cumini Linn.) Terhadap Bakteri Shigella dysentriae Secara In Vitro

Abstract

Potensi tanaman herbal berkhasiat banyak dimanfaatkan untuk mengobati berbagai macam penyakit. Daun juwet (Syzygium cumini Linn.) secara tradisional biasa digunakan dalam pengobatan disentri oleh masyarakat di Indonesia. Penelitian ini bertujuan membuktikan adanya efek antibakteri ekstrak daun juwet terhadap Shigella dysentriae secara in vitro. Aktivitas antibakteri ekstrak daun juwet dievaluasi menggunakan metode dilusi agar. Konsentrasi ekstrak yang diujikan antara lain 0%, 0,2%, 0,3%, 0,4% dan 0,5%. Proses ekstraksi dilakukan dengan cara maserasi dalam pelarut etanol 70%. Hasil penelitian diperoleh bahwa nilai konsentrasi hambat minimum (KHM) ekstrak etanol daun juwet terhadap Shigella dysentriae adalah 0,5%. Ekstrak etanol daun juwet (Syzygium cumini Linn.) melalui uji fitokimia positif mengandung fenol, flavonoid, tanin, saponin dan alkaloid. Analisis data menggunakan uji Kruskall – Wallis yang dilanjutkan dengan post-hoc Mann Whitney menunjukkan adanya perbedaan bermakna antara pertumbuhan bakteri dengan kadar sampel uji ekstrak (Sig 0,001 < p 0,05). Korelasi Spearman mengindikasikan adanya hubungan yang erat antara besar konsentrasi ekstrak yang diberikan dengan tingkat penurunan pertumbuhan bakteri (R = -0,961; p < 0,05). Kesimpulan dari penelitian ini adalah ekstrak daun juwet (Syzygium cumini Linn.) dapat menghambat pertumbuhan bakteri Shigella dysentriae.

English Abstract

Natural remedies are widely used for treatment of diseases. Syzygium cumini leaves commonly used in Indonesia as a traditional medicine to treat dysentery. The aim of this study was to investigate antibacterial effect of Syzygium cumini leaves extract towards Shigella dysentriae in vitro. The antibacterial activity was evaluated by agar dillution method. The tested extract concentrations are 0%; 0,2%; 0,3%; 0,4% and 0,5%. Extraction was carried out by maceration with ethanol 70% as the solvent. The result showed that MIC value was 0,5%. Phytochemical screening from ethanolic extract of Syzygium cumini leaves were phenols, flavonoid, tannin, saponin and alkaloid. Data analysis using Kruskall – Wallis test followed by post-hoc Mann Whitney showed a significant difference between bacterial growth and concentrations extract tested (Sig 0,001 < p 0,05). Spearman correlation test indicated there was strong relationship between Syzygium cumini leaves extract in various concentrations tested and bacterial growth levels. The conclusion of this study suggests that ethanolic extract of Syzygium cumini leaves inhibits the growth of Shigella dysentriae
